About this file

This document is a University of New Mexico (UNM) Proposed Sole Source Purchase Form for a specialized fiber photometry system from Doric Lenses Inc. The procurement is for a one-time purchase of a custom scientific research instrument valued at $67,510, with a proposed purchase date of January 22, 2025. The system is specifically designed for neuroscience research, enabling simultaneous dual-site recording and programmable stimulation in mouse brain research, with particular emphasis on stroke neurobiology studies. The system uniquely integrates fiber photometry with laser illumination for photothrombotic stroke research, a methodology first developed by Dr. Andrew Boyce.

The sole source justification is based on the system's proprietary technology and unique compatibility with Dr. Boyce's research approach. Doric Lenses Inc. was selected after a due diligence process that included reviewing alternative vendors like Bruker and Tucker Davis Technologies, which were determined to be less suitable for the specific research requirements. The DANSE analysis software accompanying the system requires a yearly $2,000 subscription. The procurement follows New Mexico Procurement law, which mandates posting the form on the UNM Sunshine Portal and State of New Mexico Procurement website for 30 days prior to purchase, ensuring transparency in the sole source acquisition process.

One Time Purchase

1/22/2025 196666175 http://purchase.unm.edu/department-information/comm-code-listing.pdf Andrew Boyce A fibre photometry system allows detection of changes in genetically-encoded fluorescent biosensors (sensitive to important signalling molecules, neurotransmitters, etc.) expressed in the brain of a freely behaving mouse. Recording of these events through a chronically implanted optical fibre, attached to the system through a fibreoptic patchcord, allows for simultaneous detection of cell- and region-specific changes in brain activity with unimpeded mouse behaviour. This particular system allows for dual site recording with concurrent programmable stimulation in two animals simultaneously. This custom Doric system is “plug and play” through the whole process, including both data collection and analysis platforms - enhancing usability.

Further, this system is fully integratable with a laser illumination that is required for induction of photothrombotic stroke. The combination of these two approaches was first developed by Dr. Boyce on this Doric system and has yet to be demonstrated on any other system.

Andrew Boyce A fibre photometry system (paired with stroke induction) will be an integral part of the new lab of Dr. Boyce being established in the Neurosciences Department. The plan is to be using this system at least 5 days per week to collect important data for the growth of his lab and the study of stroke neurobiology.

Andrew Boyce As mentioned above, this system is fully integratable with a laser illumination that is required for induction of photothrombotic stroke. The combination of these two approaches was first developed by Dr. Boyce on this Doric system and has yet to be demonstrated on any other system.

This particular system will allow us to record in dual colour (ie. two independent biosensors for distinct signalling molecules) from multiple animals simultaneously, allowing an understanding of interactions at the cell signalling as well as the whole animal level.

Further, this system uses the DANSE analysis software that has been developed in parallel with the development of the combination photothrombotic stroke/fibre photometry by Dr. Boyce.

III. SUPPLEMENTAL DETAILS. PLEASE PROVIDE ADDITIONAL INFORMATION AS

REQUESTED BELOW:

Describe in detail the unique capabilities of the proposed vendor’s goods/service and/or personnel performing the work and why this constitutes the only source. Focus on what is unique about the goods/service and why no other vendor could meet your needs.

Andrew Boyce The DANSE analysis software license requires a yearly subscription of $2000 USD to maintain, otherwise service requirements are minimal and damaged equipment can be replaced part-wise if necessary.

Andrew Boyce The CBRR core has multiple Doric miniaturized microscopes that run on the same data collection platform and could be integrated with the purchased fibre photometry setup.

Andrew Boyce As mentioned above, this system is fully integratable with a laser illumination that is required for induction of photothrombotic stroke. The combination of these two approaches was first developed by Dr. Boyce on this Doric system and has yet to be demonstrated on any other system.

The Doric system also offers more flexibility for in vivo electrophysiology integration (particularly for long term recordings with integrated rotary joint) to allow detection of post-stroke seizure in mice.

Further, this system uses the DANSE analysis software that has been developed in parallel with the development of the combined photothrombotic stroke/fibre photometry by Dr. Boyce.

Describe the due diligence made to locate other possible sources including communications with other universities, communications with similar providers, web searches, yellow page searches, review of advertisements and trade publications, etc.

List the other vendors who were contacted. Please describe the specs/qualifications/criteria that the other vendors were unable to satisfy.

During the initial establishment of this model, as well as throughout my postdoctoral training and in the past months following my hire at the UNM, I was in dialogue with several other companies’ reps about their fibre photometry systems (email correspondence, conference interactions, visits). Most recently, at SfN I visited several other booths and met with reps from other companies to understand the latest innovations in their approaches. There, I was able to review the advertisements and trade publications, as well as talk directly with their sales teams and application specialists.

Andrew Boyce Bruker - Chromatone system - while this system allowed multisite recordings, there was no sufficient way to integrate the photothrombotic stroke illumination at a sufficient power level to induce the stroke through the same optical implants.

Tucker Davis Technologies - Fibre Photometry system - The Tucker Davis system has less flexibility with regards to integration of in vivo electrophysiology (paired with photometry and stroke induction).
