About this file

This document is a Justification for Other than Full and Open Competition for reagents to support ongoing Next Generation Sequencing (NGS) projects at the National Institutes of Health (NIH), specifically the National Institute of Allergy and Infectious Diseases (NIAID). The Research Technologies Branch Genomics Research Section requires 10x Genomics single cell sequencing reagents to conduct research for NIAID investigators, including 6 units of GEM-X Universal 3' Gene Expression v4, 4 units of GEM-X Universal 5' Gene Expression v3, and multiple other specialized kits for single cell RNA sequencing, TCR/BCR sequencing, and cell surface protein expression analysis. The justification cites 41 USC 253(c)(1) for a brand-name requirement, noting that 10x Genomics offers unique proprietary microfluidic circuits and GemCode Technology that cannot be replicated by other vendors, and changing platforms would require approximately one year of work and compromise research continuity.

Justification for Other than Full and Open Competition at or Below the Simplified Acquisition Threshold

Project Title: Reagents for ongoing NGS projects

Identification of the agency and contracting activity

DHHS, NIH, NIAID, RTB, GRS

Genomics Research Section

10 Center Drive Bldg 10, Room 75N109, Bethesda, MD 20892

Background and Justification

A. Project Background

The Research Technologies Branch provides intramural scientists with state-of-the-art facilities and expertise in flow cytometry, microscopy, and genomics. The RTB Genomics Research Section (GRS) regularly performs state of the art research for clinicians and researchers within the DIR towards the NIAID goal of the discovery of new diagnostics, therapeutics, and vaccines against human disease. The reagents in this order are required for single cell sequencing of RNA. More specifically, this research makes use of 10x Genomics single cell platform to interrogate gene expression at the single cell level along with cell surface protein expression and TCR and BCR VDJ sequencing. All 10x Genomics single cell libraries are sequencing ready for Illumina NGS platforms.

B. Project Description

The Genomics Research Section currently uses 10x Genomics technologies to conduct experiments within the lab every day on behalf of all the NIAID DIR scientists. This acquisition is required to obtain the essential reagents to conduct the research projects for NIAID Investigators.

Statutory authority permitting other than full and open competition

☒ 41 USC 253(c)(1) and FAR Part 6.302-1: Brand Name Requirement

Demonstration that the proposed brand name product’s salient characteristics (specific features, qualities and/or the nature of the acquisition) requires use of the authority cited above:

10xGenomics (Manufacturer) offers the required proprietary microfluidic circuits that rapidly partition input samples (e.g. higher molecular weight DNA, single cells), which are then barcoded using the GemCode Technology.

This technology is not manufactured by any other vendor than 10X Genomics is not available for any other instrumentation. The requested reagents meet our need to sequence thousands of transcriptomes in a single run from single cells and while obtaining phenotypic data from those same cells. The latter is performed by CITE-seq, which has only been tested on the 10X platform; changing the platform would require about a year of work, be costly and results could not be adequately compared to data from previous studies.

Requirements for the reagent kits:

• Simultaneously measure cell surface protein expression with gene expression

• Simultaneously measure TCR, and 3’ gene expression in the same cells

• Be able to detect protein of low abundance transcripts with increased phenotypic specificity

• Allow cell phenoptyping with hashtag technology that should also allow combining different donor samples

• Allow subcellular gene signature technology probe-based

• The kit must be compatible with the controller

Determination by the Contracting Officer that the anticipated cost/price to the Government will be fair and reasonable.

Certification This is to certify that the justification for the proposed acquisition has been reviewed and that to the best of my knowledge and belief the information and/or data provided to support my request (for the requestor) and approval (for the CO) of is accurate and complete.
