About this file
This document is a Justification for Other than Full and Open Competition for reagents to support spatial transcriptomic research of Human Norovirus. The Vaccine Research Center (VRC) at the National Institutes of Health (NIH) is seeking to purchase specialized reagent kits from 10X Genomics to study host genes involved in norovirus infection. The specific products include Xenium Standalone Custom Gene Panels, Xenium Advanced Panel Upgrades, Cell Segmentation Add-on Kits, Decoding Consumables, Decoding Reagents, and Slides & Sample Prep Reagents, with quantities ranging from 1 to 8 units.
The justification emphasizes that 10X Genomics is the sole provider of these proprietary microfluidic circuits and GemCode Technology, which enables sequencing thousands of transcriptomes in a single run while obtaining phenotypic data. The reagent kits must meet specific requirements, including simultaneously measuring cell surface protein expression, TCR, and gene expression, detecting low abundance transcripts, and being compatible with the controller. The procurement is authorized under 41 USC 253(c)(1) and FAR Part 6.302-1 for brand name requirements, with Contracting Officer Addison Miller approving the acquisition on 9/11/2025.

Justification for Other than Full and Open Competition Brand Name at or Below the Simplified Acquisition Threshold
Project Title: Reagents for Spatial Transcriptomic of Human Norovirus
Identification of the agency and contracting activity
DHHS, NIH, NIAID, VRC
Vaccine Research Center 40 Convent Drive, Bldg. 40, Room 3614, Bethesda, MD 20892
Background and Justification
A. Project Background
Vaccine Research Center (VRC) mission is to discover and develop novel vaccines and biologics targeting infectious diseases of global public health importance. VRC combines a scientific and technological strategy innovation to achieve the design, development, manufacturer and evaluation of vaccines and therapeutics. Tissue Analysis Core
(TAC) contributes to VRC mission by characterizing immune signatures at tissue level to better understand natural infection and strategies of intervention and vaccines.
B. Project Description
To date little is known about host genes required for HNoV infection, cell tropism, immune signatures and immune evasion. Spatial transcriptomic has been determined to be the best approach to profile the cellular heterogeneity of tissues like stomach and intestine during HnoV infection. These supplies will be used in support of those research studies.

Statutory authority permitting other than full and open competition
☒ 41 USC 253(c)(1) and FAR Part 6.302-1: Brand Name Requirement
Demonstration that the proposed brand name product’s salient characteristics (specific features, qualities and/or the nature of the acquisition) requires use of the authority cited above:
10xGenomics (Manufacturer) offers the required proprietary microfluidic circuits that rapidly partition input samples (e.g. higher molecular weight DNA, single cells), which are then barcoded using the GemCode Technology.
This technology is not manufactured by any other vendor than 10X Genomics is not available for any other instrumentation. The requested reagents meet our need to sequence thousands of transcriptomes in a single run from single cells and while obtaining phenotypic data from those same cells. The latter is performed by CITE-seq, which has only been tested on the 10X platform; changing the platform would require about a year of work, be costly and results could not be adequately compared to data from previous studies.
Requirements for the reagent kits:
• Simultaneously measure cell surface protein expression with gene expression
• Simultaneously measure TCR, and 3’ gene expression in the same cells
• Be able to detect protein of low abundance transcripts with increased phenotypic specificity
• Allow cell phenoptyping with hashtag technology that should also allow combining different donor samples
• Allow subcellular gene signature technology probe-based
• The kit must be compatible with the controller
Determination by the Contracting Officer that the anticipated cost/price to the Government will be fair and reasonable.
Certification This is to certify that the justification for the proposed acquisition has been reviewed and that to the best of my knowledge and belief the information and/or data provided to support my request (for the requestor) and approval (for the CO) of is accurate and complete.
