The Proteins & Chemistry Section (PCS) provides support for planning and executing proteomics, metabolomics, lipidomic, and bioactive small molecule studies for intramural investigators. This includes assistance with purification, identification, and quantitative analysis using primarily mass spectrometry-based approaches. The section supports all facets of these projects, including but not limited to project design, separation protocols, sample preparation, chemical labeling for quantitative proteomics, derivatization for improved coverage in metabolomics workflows, enrichment for post-translational modification analysis, and postacquisition data analysis. In the fall of 2025, the NIH Standardized Organoid Modeling (SOM) Center was launched with the aim of coordinating existing expertise across the NIH in order to establish best practices and analytical tools for better calibration of organoid models. As part of the SOM Center initiative, the Protein Chemistry section is expanding its instrumentation to provide access to high-sensitivity single-cell proteomics resources capable of deep visual proteomics, high-throughput post-translational modification analysis, and protein/peptide quantitation. The lmd7 has a unique gravity assisted collection method that avoids manual manipulation of the dissected piece reducing sources of contamination that can add unacceptable noises to the organoid workflow. No other NIH mass spectrometry group supports deep visual proteomics and attempts to collaborate with outside HHS has failed due to the complexity of working with biological systems.
