Justification for Other than Full and Open Competition at or Below the Simplified Acquisition Threshold
Project Title: Acquisition of 10X Genomics Library Construction Kits and associated components
I . Identification of the agency and contracting activity
DHHS, NIH, NIAID, LISB
Requestors name: Rachel Sparks Requestors phone number: 240-292-4642 Requestors email address: rachel.sparks@nih.gov Address of the requesting lab/office: 4 Memorial Dr.
Building 4 Room 138 Bethesda, MD 20892
II. Description of the project, and supplies or services required to meet the agency’s needs (including the estimated value) A. Type of action. Check the appropriate box:
☒ New requirement ☐ Follow-on
• Name of the previous contractor(s): ___________________________
• Sequence number(s), if available: ___________________________________
• Was the initial and any subsequent awards competed? ☐ Yes ☐ No
B. AMBIS Sequence number for this requirement: 2175557 C. Total estimated dollar value of the requirement (including options, if applicable): $ 130,852.50 D. Period of performance, inclusive of options: Indefinite E. Project Background:
The LISB is studying the response of the immune system to the SARS-CoV-2 virus at single cell resolution. The 10X Genomics single cell platform allows genomic profiling of tens of thousands of single cells to see how proteins regulate gene expression at high resolution in an easy-to-use platform, consisting of the 10X Chromium Controller and Chromium X hardware, and the companion kits of reagents and consumables that allow use of the system. To help understand the host immune response to SARS-CoV-2 infection, the Sparks Lab is studying how the immune system responds differently for individuals who have recovered from COVID- 19, including in response to vaccination with the seasonal influenza vaccine. This is of critical importance given the global burden of SARS-CoV-2 infection and the as-of-yet incomplete understanding of how this virus impacts the immune system long-term.
Project Description:
These reagents will be used on an existing NIH-owned 10x Chromium Controller and Chromium X to examine the transcriptome state of millions of blood immune cells from COVID-19-recovered individuals. Chromium Single Cell Immune Profiling allows us a multiomic solution such as analysis of full-length V(D)J sequences for paired B-cell or T-cell receptors, cell surface protein expression, antigen specificity, and/or CRISPR edits, along with gene expression, all from a single cell. These data will allow the dissection of cell types, cell states, and gene-gene networks that underly immune responses in health and disease. The single cell profiling assays and associated libraries that will be generated as part of this project include CITE-seq, ATAC-seq, and fixed cell transcriptional profiling.
III. Identification of the statutory authority permitting other than full and open competition (SELECT ONE) ☒ 41 USC 253(c)(1) a n d F A R P a r t 6 . 3 0 2 - 1 : Only one responsible source ☐ 41 USC 501 a n d F A R P a r t 8 . 4 0 5 - 6 : Federal Supply Schedule (FSS), limited competition
☐ An urgent and compelling need exists, and following the procedures would result in unacceptable delays
☐ Only one source is capable of providing the supplies or services required at the level of quality required because the supplies or services are unique or highly specialized; or
☐ In the interest of economy and efficiency, the new work is a logical follow-on to an original Federal Supply Schedule order provided that the original order was placed in accordance with the applicable Federal Supply Schedule ordering procedures. The original order or BPA must not have been previously issued under sole-source or limited-sources procedures.
☐ 41 USC 253(c)(2) a n d F A R P a r t 6 . 3 0 2 - 2 : Unusual & compelling urgency ☐ 41 USC 253(c)(3) a n d F A R P a r t 6 . 3 0 2 - 3 : Industrial mobilization; or engineering, developmental, or research capability; or expert services ☐ 41 USC 253(c)(4) a n d F A R P a r t 6 . 3 0 2 - 4 : International agreement ☐ 41 USC 253(c)(5) a n d F A R P a r t 6 . 3 0 2 - 5 : Authorized or required by statute ☐ 41 USC 253(c)(6) a n d F A R P a r t 6 . 3 0 2 - 6 : National security ☐ 41 USC 253(c)(7) a n d F A R P a r t 6 . 3 0 2 - 7 : Public interest
IV. Demonstration that the proposed contractor(s) unique qualifications or the nature of the acquisition requires use of the authority cited.
A. Name and address of the proposed contractor(s):
10X Genomics, Inc.
7068 Koll Center Parkway Suite 401 Pleasanton, California 94566 United States
B. Describe why only the requested supply/service will meet the government’s needs:
The Chromium kits and reagents are specific to the 10X Chromium Controller and Chromium X available for our use; no other kits/reagents are compatible with these instruments, and attempting to run third party reagents would not be supported by the instrument manufacturer.
C. Describe why only the proposed contractor is capable of providing the requested supply/service:
The 10X Chromium Controller and Chromium X systems require proprietary reagents and consummables that can only be provide by the manufacterer, 10X Genomics.
V . Determination by the Contracting Officer that the anticipated cost/price to the Government will be fair and reasonable.
The price is determined to be fair and reasonable based on (provide a narrative describing how the determination was made and the rational for the determination):
VI. Certification This is to certify that the justification for the proposed acquisition has been reviewed and that to the best of my knowledge and belief the information and/or data provided to support my request (for the requestor) and approval (for the CO) of is accurate and complete.
