About this file

This document is a Single Source Determination using Simplified Acquisition Procedures (SAP) for a federal contract opportunity not exceeding the Simplified Acquisition Threshold.

The National Marine Fisheries Service (NMFS) and Southwest Fisheries Science Center (SWFSC) require whole genome resequencing of leatherback DNA to generate high-quality genome sequence data to characterize genomic diversity and investigate population genomics for leatherback turtles, as required under the Endangered Species Act. The contract is a firm-fixed price with a period of performance from September 25, 2024 to September 24, 2026, including a one-year option period. The government has determined that Novogene Corporation is the only vendor capable of meeting the minimum data quality requirements and efficient turnaround time, as they are the only company with the Falcon automated workflow system to ensure consistent, high-quality genome sequencing data. No other vendors were found capable of performing the required services.

SINGLE SOURCE DETERMINATION USING

SIMPLIFIED ACQUISITION PROCEDURES (SAP)

FOR AN ACTION NOT EXCEEDING THE SIMPLIFIED ACQUISITION THRESHOLD

(AUTHORITY: FAR 13.106-1(b)(1))

143051-24-0105

1. Agency and contracting activity. Department of Commerce, NOAA Acquisition and Grants Office (AGO), National Oceanic Atmospheric Administration (NOAA), National Marine Fisheries Service (NMFS) and Southwest Fisheries Science Center (SWFSC), Marine Mammal and Sea Turtle Division (MMTD).

2. Description of supplies or services required to meet agency needs (including the estimated value).

SWFSC requires whole genome resequencing of leatherback DNA to generate whole genome sequence data of sufficient quality to characterize genomic diversity and investigate aspects of population genomics for leatherback turtles (justified under the requirements of the Endangered Species Act). Delivery of the sequence data is expected approximately 21 days after receipt and quality verification of leatherback DNA samples, and service includes all DNA quality checks, quantification, sequence library preparation, pre-made library sequencing, sequence data generation and final data filtering/quality assessment.

Period of Performance: September 25, 2024 – September, 24 2026 (Base: 9/25/24-9/24/25; plus 1 option year: Sept. 25, 2025-September 24, 2026) Expected delivery date: Within 2-3 weeks of receiving the DNA samples from SWFSC

Contract Type: Firm Fixed Price

3. Identification of the single source or the brand name to be solicited.

Vendor name: Novogene, Address: 2921 Stockton Blvd. Suite 1810, Sacramento CA 95817 Lucy Keegan 443-822-7524

4. Supporting rationale. Only one source or brand name is reasonably available as detailed below:

Other - Only one source is capable -

Novogene is largest sequencing center in the world, housing a full selection of Next- Generation sequencing platforms, including the (here at their California facility) which ensures they can generate whole genome sequence data of sufficient quality and efficient turnaround time to meet the requirements for this project. They have experience sequencing leatherback and other sea turtle DNA for similar projects worldwide, and thus are able to facilitate coordination among sea turtle genome researchers to technical specifications established by the Working Group.

Novogene is

143051-24-0105

Source Selection Information – see FAR 2.101 and 3.104 the only company running the Falcon automated workflow software system allowing fast turnaround time and higher data quality guarantee (for a lower price) than other companies. They are thus able to meet our minimum data quality guarantee (30G data per sample, Q30 85%). These projects require consistent data quality and coverage methods in order to provide quality control and comparability among genome sequences. Failure to meet these requirements will result in incompatibility with existing published genomes for comparison, and unacceptable error rates in genome sequences.

5. Market Research.

Commercial whole genome sequencing has primarily been developed to meet the human, biomedical and agricultural needs. For endangered species, it remains a research-driven process, primarily conducted through large genomics centers at universities. While this industry is evolving rapidly as larger companies acquire start-ups and emerging technologies, Novogene is the only company to have developed an automated pipeline of the selected options of sample prep and sequencing technologies to be able to guarantee our minimum data quality standards in the required turn-around time for the scale of our project. After thorough market research, no other vendors are capable of performing the specific tasks needed for our project.

6. Technical/Requirements Representative Certification

I certify that this requirement constitutes the Government’s minimum needs and the supporting data provided herein is accurate and complete to the best of my knowledge and belief.

7. Determination

I hereby determine that the circumstances of this action deem only one source is reasonably available. This determination is accurate and complete to the best of my knowledge and belief.
