Bureau of Land Management, Rawlins Field Office Statement of Work Muddy Creek Aspen Genetics
1.0. SCOPE OF WORK
The Contractor will determine clonal membership, cytotype, and sex for 700 individual leaf samples collected over landscapes containing isolated aspen stands in southern Wyoming. Leaf samples will be collected by the Bureau of Land Management Rawlins Field Office and provided to the contractor Fall 2024 and Fall 2025. The contractor will then perform the DNA extractions and genotyping. Once DNA has been extracted from all samples, Contractor will perform a reduced-representation sequencing analysis of all samples (i.e. RADseq) following Blonder, Walton & Mock (2020). Based on genomic analyses in aspen, we anticipate that the RADseq sequencing will yield many tens of thousands of single nucleotide polymorphisms in each sample, and that the sequencing depth will be sufficient for robust determination of clonal membership and genome-wide ploidy levels (Gompert & Mock 2017; Blonder et al. 2021; Goessen et al. 2022; Blonder, Walton and Mock 2020; Blonder et al. 2023). Sex will be assigned using the TOZ19 male-specific gene (Pakull et al., 2015), which is reliable in Populus (Pakull et al., 2011) and in diploid aspen (Bidner, 2021).
Results, including interpretation and recommendations, will be provided in the form of a report with open source links to raw and processed data and bioinformatic scripts.
2.0. PERFORMANCE SPECIFICS
Place of Performance: The Place of Performance will be the contractor's location.
Period of Performance: October 1, 2024 - July 31, 2026 This would be split into two periods. First, October 2024 - July 2025 for 200 sample; and second, October 2025 - July 2026 for 500 samples.
Material will be collected and available to send by:
October (Fall) 2024 - Material (200 Leaf samples) October (Fall) 2025 - Material (500 Leaf samples)
Effort: The Contracting Officer's Representative (COR) must be apprised of work schedules.
3.0. BACKGROUND:
The Bureau of Land Management (BLM) High Desert District Rawlins Field Office is seeking genetic testing on Aspen within the Muddy Creek Focal Area. Aspen (Populus tremuloides) is a common forest tree species in North America. This species is particularly important in landscapes of the Intermountain West, where striking mortality and stand loss has been noted in recent decades due to drought and ungulate browsing driving regeneration failure (Anderegg et al. 2013, Dudley et al. 2015). A common pattern of aspen occurrence in western sage steppe landscapes is in isolated swales and drainages, which may be remnants of a much more broadly distributed aspen forest type. These isolated stands can be especially valuable as wildlife habitat and livestock shelter, and are frequently pockets of diversity for plants, birds, and insects. Because of their isolation, however, these stands can be particularly vulnerable to losses.
Individual aspen clones can be quite large and thousands of years old (Ally et al. 2010), raising the question of whether over long time spans clones can become fragmented into different stands across a landscape, reducing regional diversity. Similarly, single clones can dominate small stands over time, and these more dominant clones are frequently triploids (Mock et al. 2008, 2012). Dominance by triploids may be problematic because triploids are expected to have reduced fertility, meaning that the probability of local post-disturbance sexual reproduction (e.g. after fire) may be greatly reduced. Another issue in small stands is that one sex (especially males) can come to dominate the stand, which would also decrease the probability of post-disturbance sexual reproduction (Bidner 2021). Overall, with changing climates, sexual reproduction in aspen will be a key factor in adaptation and persistence (Long & Mock 2012). Therefore, understanding the clonal, cytotype (ploidy level), and sex composition of small stands is critically important for the management and restoration of these stands into the future.
Area of Interest Rawlins Filed Office Muddy Creek Watershed Focal Area, Aspen Stands.
4.0. TASKS
The primary scope of this requirement, the Contractor shall conduct the following tasks/work:
· 700 DNA extractions on leaf samples obtained by the Field Office
· Reduced-representation sequencing analysis of all samples (i.e. RADseq.) following Blonder, Walton & Mock (2020)
· Determine of clonal membership and genome-wide ploidy levels (Gompert & Mock 2017; Blonder et al. 2021; Goessen et al. 2022; Blonder, Walton and Mock 2020; Blonder et al. 2023)Participate in the monthly/weekly BLM conference calls and in-person meetings.
· Assign sex using the TOZ19 male-specific gene (Pakull et al., 2015)
· Prepare monthly status reporting in a format agreed upon by the Contract Officer's Representative
(COR).
5.0. DELIVERABLES
The Contractor shall deliver:
Results, including interpretation and recommendations, will be provided in the form of a report with open source links to raw and processed data and bioinformatic scripts. Report will be completed within 9 months of receipt of the complete sample set.
All deliverables will reside in the public domain and remain the property of the BLM. Data will be delivered on standard external hard drive or other media with prior approval by the BLM. All data will be compatible. All deliverables will be inspected for content, completeness, accuracy, and conformance to the order requirements by the COR or task manager.
6.0. INVOICING
The Firm-Fixed price amount can be billed once deliverables are accepted this can be done in one final billing or in two split invoices by year sampling. All Bureau of Land Management invoices will be submitted in accordance with the DOI Electronic Invoicing and Payment Requirements - Invoice Processing Platform (IPP).
7.0. GOVERNMENT FURNISHED ITEMS
· Any necessary data as identified during the project will be provided to the contractor on the proper and agreed upon media.
