1. For amplicon project:
a. Which region would you like to analyze, i.e. V3-V4 region of 16S rRNA gene?
Ans: mostly V3-V4, but other regions may also be targeted.
b. What's the sequencing configuration are you going to use (i.e. sequencing platform, reads number per sample)?
Ans: Sequencing will be performed on Illumina MiSeq using 300 bp paired-end
MiSeq protocols, reads number varies according to sample.
2. For Whole Metagenome Shotgun sequencing project:
a. What's the sequencing configuration are you going to use (i.e. sequencing platform, data output per sample)?
Ans: Oxford Nanopore typically with 40 million reads minimum
3. For Whole Genome Sequencing project:
b. a) What organism are you working with?
Ans: Several bacterial and viral species
c. b) What is the (estimated) genome size of that organism? Is a reference genome available?
Ans: These are environmental microbes with no good data on genome size and no reference genome in most cases other than those from related species.
d. Which type of service does the data come from? Whole genome de novo sequencing by the third generation sequencing technology or whole genome re-sequencing by
Illumina?
Ans: Either or both Nanopore Illumina MiSeq
