Questions & Answer
1) Is USFWS going to provide the Dart panel to the tender's winner?
The tender’s winner will be provided with the pre-developed SNP loci, age and sex panels.
2) Is it possible to get a preview/template of the panel, or alternatively know what file format they're using?
The SNP information will include the ~ 65bp allele sequence and SNP position/nucleotide for each SNP locus.
3) Is the 3000 SNPs panel in development, developed or to be developed by the tenderer?
The 3000 SNP panel has been developed using DArTseq platform and will be shared with the tender’s winner. If the tender’s winner is unable for any reason to use a pre-developed marker, they will have to develop a new SNP panel with the same number of informative loci to be used in the sample processing.
4) Are samples going to be delivered as data and/or biological material? In the latter case, how is nucleic acid extraction going to be performed?
The samples are going to be delivered as biological material (skin) plated/stored according to the vendor’s technical requirements. The vendor is expected to perform DNA extraction from the provided samples using any available extraction methods allowing to obtain high purity DNA suitable for further genotyping.
5) Is the tenderer supposed to use only Dart technology or can it use its own resources, pipelines and technologies alternatively or in association with Dart (as long as it doesn't change it)?
The tender’s winner can use DaRT or any other technology allowing for the delivery of required genotyping products with negligible error rate and within identified timeframe.
6) Are Dart's data raw or already interpreted? In case the SNPs are going to be shared?
The pre-developed SNP panel will be shared with the tender’s winner.
7) Are the pipelines the tenderer is asked to develop (mt-DNA, age, sex, close-kin panels) going to be a parallel, replace or work with Dart panels?
The tender’s winner is expected to use the pre-developed SNP loci, age and sex panels to process the samples. The mtDNA marker will have to be developed from scratch. However, if the tender’s winner is unable for any reason to use any of the pre-developed markers, they will have to develop new marker(s) to be used in the sample processing.
8) When referring to plates in phase 3 and 4, does USFWS mean one sample per plate or one sample per well?
“Plate” refers to a standard PCR plate with 96 wells (94 samples plus 2 negative controls per plate).
