STATEMENT OF WORK
TITLE: Acquisition of High Resolution Mass Spectrometry Instrument
LAB REQUESTING SERVICE: Materials Measurement Laboratory, Materials Measurement Science Division
I. BACKGROUND INFORMATION
The National Institute of Standards and Technology’s Chemical Sciences Division serves as the Nation's reference laboratory for chemical measurements and standards to enhance U.S. industry's productivity and competitiveness, assure equity in trade, and provide quality assurance for chemical measurements used for assessing and improving public health, safety, and the environment. The Chemical Sciences Division is responsible for development and characterization of Standard Reference Materials (SRMs) to enhance chemical measurement comparability within the U.S. and to provide a basis for measurement comparability with other nations. SRMs are provided to support clinical, food, environmental, fuel, metal alloy, cement, and advanced materials analyses. Research activities are geared toward improvement of measurement capabilities to address U.S. industry and government agencies' requirements for new measurement services and reference materials. Current research focus areas include the development of measurements and standards for determination of protein-based health markers, chemical characterization of complex matrices (e.g. lipidomics and metabolomics), and for environmental contaminant analysis.
The Chemical Services Division maintains core expertise in:
• Analytical mass spectrometry,
• Analytical separation science,
• Atomic and x-ray fluorescence spectrometry,
• Gas metrology,
• Nuclear analytical methods,
• Nuclear magnetic resonance spectrometry, and
• Classical and electroanalytical chemistry.
Because of NIST’s role as the National Metrology Institute (NMI) in the U.S., the Division’s measurement capabilities must be benchmarked against the capabilities of NMIs worldwide as required by a Mutual Recognition Arrangement (MRA) established in 1999. The MRA provides a framework for obtaining reliable quantitative information on the comparability of measurement services provided by all NMIs, thereby providing governments and other parties with a technical foundation for wider agreements related to international trade, commerce, and regulatory affairs.
The Division actively participates in the International Committee of Weights and Measures (CIPM) Consultative Committee for Amount of Substance – Metrology in Chemistry (CCQM) comparisons, which address chemical measurement-related issues important for international trade, environmental, health, and safety-related decision making.
II. SCOPE OF WORK
NIST intends to acquire a high resolution mass spectrometer used for the metabolite and lipid profiling, identification of proteins by intact mass analysis, peptide mass fingerprinting, identification and structural elucidation of posttranslational modification (e.g. phosphorylation and glycosylation), and the quantification of peptides, proteins, lipids, and metabolites. The instrument must be able to reproducibly generate high resolution and high-quality MS and MS/MS in a variety of fragmentation modes in order to accurately identify unknown analytes in complex matrices.
This instrument must also give reproducible quantitative results compared to other MS/MS instruments.
The required equipment is specifically designed for high resolution mass spectrometric measurements. The required equipment consists of benchtop instruments with integrated instrument control and data analysis software. The inclusion of an HPLC or UPLC is not required, however the mass spectrometer instrument and software must be capable of utilizing, with full software seamless integration and automation, existing LC equipment specifically a Thermo Vanquish UPLC and a Dionex Ultimate 3000 UPLC.
III. SPECIFICATIONS
This is a brand name or equal requirement. Brand name or equal description is intended to be descriptive, but not restrictive and is to indicate the quality and characteristic of products that will be considered satisfactory to meet the agency’s requirement.
The Contractor shall provide the following Thermo Electron branded equipment or equivalent, and software for complete instrument system control and data analysis capabilities meeting the salient characteristics as defined below:
A. One (1) Thermo Electron Q Exactive High Resolution Mass Spectrometer (MS) with Ion Max Source and Heated Electro Spray Ionization (HESI-II) Probe (Mfr. Part Number 0726020) and the Exactive Series Pre-Installation Kit (Mfr. Part Number 1328020) or a brand with the same salient physical, functional and performance characteristics set forth below. The system shall meet or exceed the technical specifications identified below and shall be brand new (prototypes, demonstration models, used or remanufactured equipment will not be considered for award):
a. High resolution orbitrap mass spectrometer with quadrupole mass filter (capable of parallel filling to increase instrument duty cycle)

c. Resolution - 130,000 (FWHM) @ 200 m/z 40,000 (FWHM) @ 1000 m/z
d. Mass range - 50 – 4000 Da
e. Dynamic range (full MS scan) – 5,000
f. Sensitivity (signal to noise) - 100:1 of 500 fg reserpine (Full MS)
g. Mass accuracy - 5 ppm (external) 1 ppm (internal calibration)
h. Polarity switching - 1 sec between positive and negative scans (with same mass resolution and mass accuracy performance as above)
i. MS/MS Fragmentation - higher energy collisional dissociation (HCD)
j. Scan functions must include:
Full MS Selected ion monitoring (SIM)
Selected reaction monitoring (SRM) Multiplexed SRM Multiplexed SIM Data dependent analysis (DDA) Data Independent Analysis (DIA) Neutron Loss Triggered DDA Timed SIM Timed SRM Dynamic exclusion
k. HESI/ESI Source operating voltage - up to ± 6000 V (100 µamps)
l. HESI/ESI Liquid flow rate operation range - 1 µL/min – 1000 µL/min
m. HESI/ESI Temperature up to 400 ºC
The instrument software must be compatible with Windows 7 operating systems. The instrument software must be able to control and seamlessly integrate all of the MS and LC functionality (i.e. existing Thermo Vanquish UPLC and a Dionex Ultimate 3000 UPLC) with a single software program/package to ensure efficient workflow and transition between systems.
Warranty:
The Contractor shall provide, at a minimum, a three-year warranty for the system’s non-consumable parts. The warranty shall cover the following:
