COMBINED SYNOPSIS/SOLICITATION
Description: Amendment—Revised Technical Specifications and Delivery Date. Purchase of an Applied Biosystems QuantStudio 3 Real-Time PCR System
(i) This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR Subpart 12.6, as supplemented with additional information included in this notice and in accordance with the simplified acquisition procedures authorized in FAR Part 13. This announcement constitutes the only solicitation; quotes are being requested and a written solicitation will not be issued.
(ii) This solicitation is issued as a request for quotation (RFQ). Submit written quotes on RFQ Number: 143071-24-0072.
(iii) The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2024-05 (May 2024).
(iv) This solicitation is being issued as Unrestricted and open to all sources. The associated NAICS code is 334516. The small business size standard is 1000 employees.
(v) This combined solicitation/synopsis is for purchase of the following commercial supply:

Purchase of Applied Biosystems QuantStudio 3 Real-Time PCR System in accordance with the Statement of Need. Deliver by November 30, 2024.

FAR 52.212-1 Instructions to Offerors-Commercial Products and Commercial Services (Sep 2023) NOTICE TO OFFERORS: Instructions for submitting quotations under this request for quote must be followed. Failure to provide all information to aid in the evaluation may be considered non-responsive. Offers that are non-responsive may be excluded from further evaluation and rejected without further notification to the offeror.

FAR 52.212-2 Evaluation- Commercial Products and Commercial Services (Nov 2021) Offers will be evaluated based on price and the factors set forth in paragraph (a).
(a) The Government will award a contract resulting from this solicitation to the responsible offeror whose offer conforming to the solicitation will be most advantageous to the Government, price and other factors considered. Paragraph (a) is hereby completed as follows: Evaluation will be based on the following;

2. Price.
The Government intends to award a low priced, technically acceptable, single firm fixed-price purchase order on an all or none basis with payment terms of Net 30.
(b) Options. The Government will evaluate offers for award purposes by adding the total price for all options to the total price for the basic requirement. The Government may determine that an offer is unacceptable if the option prices are significantly unbalanced. Evaluation of options shall not obligate the Government to exercise the option(s).
(c) A written notice of award or acceptance of an offer, mailed or otherwise furnished to the successful offeror within the time for acceptance specified in the offer, shall result in a binding contract without further action by either party. Before the offer’s specified expiration time, the Government may accept an offer (or part of an offer), whether or not there are negotiations after its receipt, unless a written notice of withdrawal is received before award.

Applied Biosystems QuantStudio 3 Real-Time PCR System
PURPOSE:
The National Oceanic and Atmospheric Administration (NOAA) Pacific Islands Fisheries Science Center (PIFSC) Ecosystem Sciences Division (ESD) seeks to purchase one a quantitative real-time PCR (qPCR) thermalcycler to support research identifying genetic material in tissues and environmental samples.
We require the purchase of the Applied Biosystems QuantStudio 3 Real-Time PCR System manufactured by Life Technologies Corporation to maintain consistency of existing qPCR projects that utilize the QuantStudio platform and reagents. The QuantStudio 3 qPCR system is a high-performance real-time quantitative PCR system with 96-well sample throughput.
BACKGROUND:
Environmental DNA (eDNA) is a powerful and emerging tool that can complement existing surveys to expand and refine our understanding of fish populations and habitat use in marine ecosystems. Currently, eDNA techniques are in the process of becoming a part of the standard fishery and wildlife management toolkit for population detection, assessment, and monitoring.
Large-scale eDNA surveys can be cost-effective ways for assessing occupancy and relative abundance of target fisheries and protected species in habitats and locations not currently surveyed by other techniques. eDNA studies commonly use quantitative polymerase chain reaction (qPCR) for the detection of low levels of target species' eDNA found in complex environmental samples such as a water sample (e.g., Shelton et al. 2022). PIFSC does not currently have access to a qPCR thermalcylcer, and thus would like to procure one to permit these types of surveys.
qPCR is one of the most common techniques for quantification of nucleic acid molecules in biological and environmental samples. In real-time PCR, the accumulation of amplification product is measured as the reaction progresses, in real time, with product quantification after each cycle. First, amplification reactions are set up with PCR reagents and unique custom-designed primers. Reactions are then run in real-time PCR instruments. Detection of PCR products is enabled by the inclusion of a fluorescent reporter molecule in each reaction that yields increased fluorescence with an increasing amount of product DNA. Fluorescently-labeled sequence-specific probes are designed to particular gene regions that selectively amplify target species. When primers and probes are designed to be species-specific, fluorescence occurs only when the forward primer, reverse primer and probe hybridize to the target species DNA.
Specialized thermal cyclers equipped with fluorescence detection modules are used to monitor the fluorescence signal as amplification occurs. The measured fluorescence is proportional to the total amount of amplicon; the change in fluorescence over time is used to calculate the amount of amplicon produced in each cycle. The main advantage of real-time PCR over PCR is that real-time PCR allows you to determine the initial number of copies of template DNA (the amplification target sequence) with accuracy and high sensitivity. Real-time PCR results can either be qualitative (the presence or absence of a sequence) or quantitative (copy number).
Quantitative real-time PCR is thus also known as qPCR analysis. Probe-based qPCR provides the opportunity to process multiple samples in a single reaction (multiplex).
To conduct large-scale eDNA surveys and have the ability to develop species-specific qPCR assays in house, PIFSC requires the procurement of a qPCR thermalcycler. We require the purchase of the Applied Biosystems QuantStudio 3 Real-Time PCR System manufactured by Life Technologies Corporation to maintain consistency of existing qPCR projects that utilize the QuantStudio platform and reagents. For example, we have developed species-specific qPCR primer-probe assays to identify Hawaiian Bottomfish from environmental samples using the Applied Biosystems QuantStudio Real-Time PCR system using TaqMan Environmental Master Mix and TaqMan multiple kits.
