About this file
This Statement of Work (SOW) outlines the USDA Agricultural Research Service Pacific Insect Research Unit's (PIRU) requirement for DNA sequencing services of 10 pooled DNA libraries using Illumina NovaSeq X Plus (PE150) sequencing technology. The SOW is part of solicitation 12905B25Q0018, issued as a total small business set-aside under NAICS 541714 with a size standard of 1,000 employees.
The contractor must sequence 10 sets of premade, pooled animal DNA libraries with Illumina-compatible dual indexing adapters, with each library requiring a full lane using a 10B flow cell to produce approximately 375G of high-quality sequence data. Each pooled library will contain 30-200 multiplexed insect samples. Requirements include quality checking before sequencing, sharing QC results with researchers, delivering data in FASTQ format via secure FTP/globus/command line, and completing each sample within one month of submission. All results must be received by May 31, 2026. Quotes are due by January 27, 2025, at 3 PM EST, with questions due by January 23, 2025, at 3 PM EST.

STATEMENT OF WORK
The USDA-ARS-PIRU has the need to sequence 10 sets of premade, pooled DNA libraries using Illumina NovaSeq X Plus (PE150) sequencing. The library type is animal, short-insert libraries with Illumina-compatible, dual indexing adapters.
Each library should be sequenced as a full lane using a 10B flow cell and each lane should produce ~375 G of high-quality sequence data.
Each premade library will contain a pooled set of insect samples that have been indexed with unique, 8bp dual-indexing adapters. These are variable, 30-200 libraries multiplexed together with unique dual indexing.
Each library needs to be quality checked (QC) (concentration and fragment distribution), prior to sequencing.
The QC results must be shared with the researchers prior to sequencing.
After sequencing, the data need to be quality checked, ensuring high-quality results. The data must be shared in FASTQ format via FTP, globus, or command line. Data must be returned via a secure website/server.
Data should be returned within one month of submission for each of the ten pooled samples.
Data can be demultiplexed or combined, with the demultiplexing done by the researchers (preferred).
Sample pools need to be submitted sequentially over the duration of the contract.
