STATEMENT OF WORK

Nitrite and Nitrate Pesticide Residues in Pig Tissues—In-Life Animal Study with Tissue Residue Analysis and Tissue Residue Storage Stability

INTRODUCTION

Within the next few years, USDA APHIS will likely be submitting a pesticide registration application to the U.S. EPA under Federal Insecticide, Fungicide, and Rodenticide Act (FIFRA; 7 USC 136) Section 3 for a product containing the unregistered active ingredient, sodium nitrite, that will be used for controlling feral swine. EPA has classified this pesticide product as having a terrestrial food use pattern (40 CFR 158). All of the applicable data requirements listed in 40 CFR 158 for a terrestrial food use product application must be met and included in USDA APHIS’ application before EPA will initiate a review.

The purpose of this work is to detect nitrite and nitrate residues in tissues from domestic pigs administered technical grade sodium nitrite by oral gavage. Specific tasks included are:

1) GLP Characterization of the test animals’ domestic pig commercial maintenance feed for nitrite and nitrate.

2) Characterization of the test animals’ drinking water for nitrite and nitrate. See section ‘LABORATORY REQUIREMENTS’ for more information.

3) Analytical method validation and characterization of test dosing solutions for sodium nitrite.

4) In-life animal phase administration of technical grade sodium nitrite by oral gavage in domestic pigs, followed by collection of muscle, adipose, liver, and small intestine tissues for analysis of nitrite and nitrate residues following EPA Residue Chemistry Test Guidelines OPPTS 860.1480 (1) as modified by the USDA-provided study design. See Attachment 1 for the study design.

5) Analysis of nitrite and nitrate tissue residues using the USDA-provided LC-UV methods for pig muscle, adipose, liver, and small intestine tissues collected during the in-life animal phase. This analysis will follow EPA Residue Chemistry Test Guidelines OPPTS 860.1480 and analytical methods provided by the USDA. See Attachment 2 for the draft analytical method conditions and extraction methods.

6) Storage stability (30-day) evaluation of nitrite and nitrate residues in pig muscle, adipose, liver, and small intestine tissues collected during the in-life animal phase per EPA Storage Stability Guidelines OPPTS 860.1380 (2).

All tasks listed above must also be conducted following EPA FIFRA regulatory requirements for Good Laboratory Practice Standards (GLPs; 40 CFR 160) (3).

DEFINITIONS

1) Federal Insecticide, Fungicide, and Rodenticide Act (FIFRA): The primary federal pesticide law (7

USC 136).

2) U.S. Environmental Protection Agency (EPA): The primary federal regulatory agency responsible for the regulation of pesticides including pesticide product registration under FIFRA.

3) Unregistered active ingredient: An active ingredient is “… an ingredient which will prevent, destroy, repel, or mitigate any pest” (7 USC 136(a) (1)). An active ingredient is unregistered when it is not already an active ingredient within currently registered pesticide products.

4) FIFRA Good Laboratory Practices (GLP): The set of organizational processes and the conditions under which non-clinical health and environmental safety studies are planned, performed, monitored, recorded, archived and reported intended to assure the quality and integrity of non-clinical laboratory studies intended to support research or marketing permits for products regulated by government agencies including the EPA (40 CFR 160) (2).

5) Institutional Animal Care and Use Committee (IACUC): The Animal Welfare Act mandates the oversight of animal welfare in federally funded animal research by IACUCs.

PURPOSE

The purpose of this contract is to fulfill data requirements for a future USDA APHIS pesticide registration application to EPA for a toxicant containing sodium nitrite for feral swine. The work will quantify the nitrite and nitrate tissue residues in domestic pigs orally exposed to the technical grade of the active ingredient, sodium nitrite. These data will be included in EPA’s human health review to evaluate potential exposure of these residues in human food.

SCOPE OF WORK

The scope of this contract will include drafting a study protocol, laboratory testing, data analysis, final report writing, and quality assurance review of the study in accordance with EPA OPPTS 860.1480 and 860.1380, and EPA FIFRA GLP requirements.

LABORATORY REQUIREMENTS

The requirements include:

1) Design and complete the study in accordance with EPA OPPTS 860.1480 and 860.1380.

Exception: The non-standard study design for the animal phase of the residue study is provided by USDA and is Attachment 1. Certain aspects of the study design are modification of the EPA OPPTS 860.1480 guidance. EPA’s concurrence with these design modifications is Attachment 3.

2) Comply with EPA FIFRA GLP requirements for all aspects of the study.

3) Draft a GLP study protocol, which will be approved by the study sponsor’s representative: USDA APHIS Wildlife Services (WS), National Wildlife Research Center (NWRC), Registration Unit (RU) prior to initiation of the study. This protocol must include all phases of the study unless a separate protocol and final report is provided for a specific component. In that event, all aspects of each study must comply with EPA FIFRA GLP requirements. Any amendments to a study protocol will be approved by the sponsor’s representative (USDA APHIS WS NWRC RU) prior to implementation. One or more aspects of the study may be subcontracted by the contracted laboratory.

4) Purchase the test substance (sodium nitrite, Sigma-Aldrich, product number S2252).

5) GLP characterization of the test animals’ domestic pig commercial maintenance feed for nitrite and nitrate.

6) The test facility must have had the test animal's supply of drinking water independently tested in a water quality analysis (e.g., an ISO 17025 Certified testing facility) within the last 12 months for both nitrites and nitrates prior to initiation of the study or must test the drinking water for these potential contaminants prior to initiation of the study.

7) GLP analytical method validation and characterization of the test substance, technical grade sodium nitrite, and test dosing solutions for sodium nitrite.

8) Conduct the in-life animal phase administration of technical grade sodium nitrite by oral gavage in domestic pigs, followed by collection of muscle, adipose, liver, and small intestine tissues for analysis of nitrite and nitrate residues following EPA Residue Chemistry Test Guidelines OPPTS

860.1480 (1) as modified by the USDA-provided study design (Attachment 1). The test animals used in this study must be from an outbred population of a common domestic pig breed (e.g., Large White F2 hybrid cross) with a known genetic background, fully weaned, and within the body size range of 18-50 kg, targeting 20-30 kg body mass. Sodium nitrite dose solution preparation and administration must occur on the same day for each test animal. Test animals that do not succumb on their own to the test substance must be euthanized immediately prior to the tissue sampling time point specified in the attached study design. Given that this study is required to determine potential tissue residues from a food use pesticide, test animals may not be euthanized once dosed and prior to the specified tissue sampling time point unless a severe injury occurs. Tissue samples must be stored as specified and sent for analytical chemistry residue analysis within 24 hours of collection.

Provide an option for an additional dose level (n = 4 test animals) if requested by the sponsor’s representative. See Price Schedule.

9) Prior to responding to this solicitation with a quote, the vendor or subcontractor for the in-life animal phase will ensure the study design of the animal phase is acceptable to the test facility

IACUC.

10) Conduct the residue analysis for nitrite and nitrate in muscle, liver, adipose, and small intestine tissues sampled during the animal phase of the residue study using analytical methods provided by the USDA and in accordance with EPA OPPTS 860.1480. Tissue residue analysis must occur within 72 hours of tissue sample collection unless prior approval has been obtained from the sponsor’s representative.

a) The draft methods are Attachment 2. The final methods are pending and will be provided by USDA prior to initiation of work.

b) The methods for all four tissues are identical with the following exceptions.

i) Liver tissue analytical method – Centrifugal separator tubes are required during extraction to minimize nitrite reactivity with endogenous proteins.

ii) Small intestine tissue analytical method – Sample dilution may be required due to potentially higher nitrite and nitrate residue concentrations.

11) Concurrently with the residue analysis, conduct a storage stability (30-day) evaluation of nitrite and nitrate residues in the pig muscle, adipose, liver, and small intestine tissues collected during the in-life animal phase per EPA Storage Stability Guidelines OPPTS 860.1380.

12) Draft a GLP final study report(s) and obtain review and approval of the final report from the sponsor’s representative: USDA APHIS WS NWRC RU prior to finalization and signing of the final report. The final report must be formatted in compliance with EPA’s PRN 2011-3: Standard Format for Data Submitted under FIFRA and Certain Provisions of FFDCA (4).

13) Test substance and sample disposal.

14) Study archiving.
