SERVICE STATEMENT OF WORK:

SECTION A - DESCRIPTION/SPECIFICATIONS/WORK STATEMENT

A.1 BACKGROUND

The USDA ARS AGIL, located at BARC-West, Beltsville, MD is conducting research on functional annotation of cattle gnome in rumen epithelial cells and to understand the development of rumen for the purposes of improving nutrition utilization efficiency.

A.2SCOPE OF WORK. The project requires:
1) ATAC-sequencing (ATAC-Seq) for 2 samples with duplicate (2X2);
2) ATAC-Seq should be optimized (ATAC-Seq optimization X 1);
3)ChIP-sequencing (ChIP-Seq) of two samples with 4 histone markers (H3K4me3, H3K27me3, H3K27ac and H3K4me1, 2X4 total 8 factorPath ChIP-seq;
4) DNA-sequencing of input/control DNA;
5) ChIP-seq with transcriptpath (CTCF ChIP-seq) (2 samples).

A.3 PROJECT OBJECTIVE

The primary objective of this work is to evevaluate the epogenomic regulation induced by nutrition element (butyrate)which is the major energy resource of cattle. The result datasets are also important for the functional annotation of cattle genome, especially the functional genomic elements that related to rumen development.
