The deliverable addresses the current limitation that existing SC-islet culture systems cannot maintain long-term viability or support fully isogenic modeling of autoimmune interactions necessary for Type 1 Diabetes research. The research product adapts a proven perfusion-capable microfluidic skin-on-chip platform into an open-well plexiglass-chamber system configured with microchannel networks to promote vascular network formation and inlet/outlet ports for perfusing endothelial cells, media, cytokines, and immune cells. The system is engineered to enable live imaging and permit removal of SC-islets and immune cells for downstream analysis. This isogenic MPS platform will support mechanistic interrogation of autoimmune pathways and cytotoxic interactions between SC-islets and autoreactive immune cells, providing a critical surrogate experimental system for understanding beta cell destruction mechanisms without requiring direct access to human pancreatic islets from living donors.Federal Project Grant Award Summary
