The $1,909,362 Project Grant awarded by the National Heart, Lung, and Blood Institute (NHLBI) under the Cardiovascular Diseases Research program (CFDA 93.837) supports the advanced preclinical development of novel NOX4 inhibitors as a therapeutic approach for pulmonary fibrosis, particularly idiopathic pulmonary fibrosis (IPF). Fibronox, LLC, a woman-owned small business, is leading this research effort in collaboration with Emory University. The goal is to conduct a rigorous evaluation of the top three NOX4 inhibitor candidates, assess their selectivity and mechanism of action, and determine the therapeutic index and maximum tolerated dose for the most promising candidate. This project builds on Fibronox's previous work identifying NOX4 as a critical mediator of lung fibrosis and demonstrating the potential of NOX4 inhibitors to protect against and potentially reverse age-associated fibrosis. The award will fund studies to further de-risk the top NOX4 inhibitor candidates, including the use of human precision-cut lung slices and single-cell RNA sequencing, to support the selection of a development candidate for IND-enabling studies and subsequent clinical development.
