The platform captures full-length proviral sequences alongside host cell transcriptomes and proteomes, preserving the natural, unstimulated state of infected cells. In Phase I, the recipient will transfer single-cell protocols to its R&D facility, validate workflows with reference standards, and establish standard operating procedures for processing blood and solid tissues including lymph nodes, brain, gut, and other clinically relevant sources. The work prioritizes access to the Last Gift cohort, a biorepository of tissues from people living with HIV who use opioids, to address an understudied population. In Phase II, the recipient will operate an in-house service laboratory to process real-world samples, enhance platform throughput and automation, refine data analysis pipelines for complex tissues and high sample volumes, and integrate improvements into a commercial-ready instrument and reagent kit.
