This Project Grant award from the National Institute on Aging (NIA), under the Aging Research Federal Grant Program (CFDA 93.866), provides $499,968.00 to Hager Biosciences, LLC to further develop novel potassium channel Kv1.3 inhibitors as potential Alzheimer's disease therapeutics. The key objectives are to: 1) complete in vitro characterization and pharmaceutical profiling of lead compounds, 2) initiate iterative lead optimization to improve potency and ADMET properties, and 3) select 2-3 lead scaffolds for further in vivo testing in rodent Alzheimer's models. This work builds on the company's prior discovery of differentiated small molecule Kv1.3 blockers, which have demonstrated efficacy in preclinical Alzheimer's models by regulating neuroinflammatory microglial activation. Successful completion of this 1-year project lays the groundwork for advancing these novel Kv1.3 inhibitors towards potential clinical development as innovative Alzheimer's disease treatments.
