The Board of Trustees of Illinois State University received a $544,734 Project Grant award from the National Institute of Arthritis and Musculoskeletal and Skin Diseases (NIAMS) under the Arthritis, Musculoskeletal and Skin Diseases Research program (CFDA 93.846), effective August 1, 2026, through July 31, 2029. This award supports research into genetic repair mechanisms and smooth muscle pathology in Duchenne muscular dystrophy (DMD), a severe genetic disorder affecting approximately 1 in 5,000 males. The project addresses a critical therapeutic gap: while existing gene therapy and corticosteroid treatments improve skeletal and cardiac muscle function, 74% of DMD patients develop life-limiting bladder dysfunction and 68% experience severe bowel complications. The research leverages C. elegans as a model organism to systematically investigate smooth muscle dysfunction across tissue types, complemented by human smooth muscle cell validation systems. Under Aim 1, the award funds comprehensive characterization of smooth muscle dysfunction using behavioral analysis, multi-compartment calcium imaging, and transcriptomic profiling. Under Aim 2, the project evaluates therapeutic efficacy of microdystrophin constructs and corticosteroids to identify differential rescue patterns across muscle types. By establishing the first cross-tissue research platform for smooth muscle DMD investigation, this work will identify tissue-specific therapeutic requirements for comprehensive treatments and provide research training for 70+ undergraduate students through direct hands-on involvement in the research program.Federal Project Grant Award Summary
