AWARD PURPOSE: THE PURPOSE OF THIS SURVEY IS TO DETERMINE WHETHER PHYTOPHTHORA RAMORUM (RAMORUM BLIGHT, SUDDEN OAK DEATH), A PATHOGEN THAT POSES A SERIOUS THREAT TO THE NURSERY INDUSTRY AS WELL AS TO THE NATURAL LANDSCAPE, IS PRESENT IN MASSACHUSETTS. TO DO THIS, WE WILL BE TARGETING HOST PLANTS FOR THIS PATHOGEN, INCLUDING SPECIES IN THESE MAJOR HOST GENERA: CAMELLIA, RHODODENDRON, VIBURNUM, PIERIS, AND KALMIA. SAMPLING WILL BE DONE AT NURSERIES THROUGHOUT THE STATE. ACTIVITIES TO BE PERFORMED: THIS SURVEY WILL FOLLOW THE CRITERIA AND PARAMETERS AS PRESCRIBED IN THE 2013 PROGRAM DIRECTED P. RAMORUM SURVEY CRITERIA, UTILIZING THE SAMPLING METHODS AND DATA COLLECTION REQUIREMENTS IN THE PHYTOPHTHORA RAMORUM NURSERY SURVEY MANUAL ON APHIS'S WEBSITE. DELIVERABLES AND EXPECTED OUTCOMES: MDAR WILL SAMPLE A MINIMUM OF 10 NURSERIES IN AT LEAST 4 COUNTIES, WITH A MAXIMUM OF 100 LEAF SAMPLES TO BE COLLECTED (APPROXIMATELY 10/SITE). LEAF SAMPLES WILL BE COLLECTED BY MDAR STAFF, STORED ACCORDING TO LAB PROTOCOLS, AND DELIVERED APPROXIMATELY WEEKLY TO THE PLANT PATHOLOGY DISEASE DIAGNOSTICS LAB AT THE PENNSYLVANIA DEPARTMENT OF AGRICULTURE (PDA-PDDL) FOR PROCESSING. PDA-PDDL WILL PROCESS THE LEAF SAMPLES USING ELISA TESTING, WITH DNA EXTRACTION AND TESTING PERFORMED FOLLOWING A POSITIVE ELISA TEST, AS PER THE NATIONAL P. RAMORUM SURVEY PROTOCOL. ANY SAMPLES THAT ARE ELISA POSITIVE WILL HAVE DNA AND TISSUE SENT TO THE USDA LAB FOR CONFIRMATION OF P. RAMORUM IN ACCORDANCE WITH THE 2020 POTENTIALLY ACTIONABLE SUSPECT SAMPLE (PASS) POLICY FOR P. RAMORUM. NURSERIES WILL BE SELECTED FOR SURVEY BY IDENTIFYING THOSE WITH THE HIGHEST RISK (THOSE WHO HAVE RECEIVED STOCK FROM STATES WHERE P. RAMORUM IS ESTABLISHED, OR FROM STATES WITH KNOWN OCCURRENCES OF P. RAMORUM), WITH PRIORITY GIVEN TO PREVIOUSLY POSITIVE NURSERIES, OR THOSE NOT RECENTLY SURVEYED FOR THIS PATHOGEN. INTENDED BENEFICIARY(IES): MASSACHUSETTS NURSERIES AND BUSINESSES WHO PRODUCE NURSERY STOCK. THE WOOD PRODUCTS AND TREE GROWERS IN MASSACHUSETTS WILL ALSO BENEFIT BECAUSE THIS WORK COULD LEAD TO EARLY DETECTION OF EXOTIC PESTS THAT MAY DAMAGE FORESTS AND TIMBER SPECIES IN THE STATE.
