Description

Purpose
The purpose of this agreement is to support research for developing resistance to the pale cyst nematode (PCN), globodera pallida, in russet type potatoes for idaho farmers who are dealing with an infestation. Potato cyst nematodes are obligate, biotrophic plant parasitic nematode, that are an economically devastating pest of potato. The quarantine potato cyst nematodes (PCN) are present in the united states, with globodera rostochiensis present in new york since 1941 and G. Pallida present in idaho since 2006. Both infestations are regulated by USDA-aphis and respective state partners because of the ability of these nematodes to significantly reduce yield of potato and because once these nematodes become established in an area IT is very difficult to eliminate them. In 2006 when PCN was detected in idaho, a major eradication effort was initiated, involving extensive use of soil fumigants. Although some resistance to PCN is present in potato varieties grown in europe, the commercial russet-skinned varieties predominating the market in idaho are susceptible to PCN. Neither resistance nor immunity traits are found in idaho's signature russet varieties. However, these traits in commercial varieties suitable for the idaho industry would be an invaluable asset in achieving the aphis goals for eradication of PCN. Financial assistance will enable the university of idaho to identify germplasm from south america (SA) for resistance and evaluate populations of G. Pallida from SA.
Activities
Identify and test potato germplasm from SA (peru, chile, bolivia, or elsewhere in SA) for resistance to potato cyst nematode (GN and PCN). Germplasms from SA have been exposed to globodera species and may have developed unique resistances. Evaluate populations of G. Pallida from SA against a set of potato differentials to characterize and evaluate different pathotypes genomes. This will help potential future introductions into the US and understand resistance to these different pathotypes. The genomes of globodera will be compared to locate differences in virulence sequences and compare the diversity of how these nematodes interact with hosts. Rearing and maintaining a population of PCN cysts. Conducting bioassays for the USDA aphis PPQ PCN program. Deliverables and expected outcomes: identification of resistant germplasms from SA to meet the goals for development of resistant potatoes suitable for idaho growers dealing with infested fields. Selection of SA PCN populations that represent a diversity of pathotypes and characterize resistance for potatoes and potential introductions into the US. PCN cyst rearing to maintain a population for testing. Conduct bioassay testing. Intended beneficiary(IES): beneficiaries include owners and operators of PCN infested fields, GN owners and operators, the potato industry in general and regulatory agencies such as aphis and the idaho state department of agriculture.
