Description
- Purpose
- Overall goal is to identify and isolate the genetic defenses from solanum sisymbriifolium, a plant known to be immune to the potato cyst nematodes (PCN), for transfer into potato. Solanum sisymbriifolium (litchi tomato-LT) is a solanaceous plant that has been shown to be nearly as effective as potato at inducing potato cyst nematode hatching but is unable to support parasitism and for this reason, has been investigated as a trap crop for G. Pallida and G. Rostochiensis. Another objective is to coordinate, and conduct the bioassay needed for DE-regulation of infested fields in a USDA-aphis-PPQ approved facility. 2.
- Activities
- A. To understand the molecular basis of plant defenses in the trap crop, solanum sisymbriifolium. Our early investigations indicated that the trap crop S. Sisymbriifolium, litchi tomato (LT) is a plant which, like potato, stimulates PCN to hatch. However, unlike potato, the plant is immune to the nematode, and PCN does not develop into cysts when infecting LT. Rather, the plant, through a hypersensitive response, walls off the nematode to prevent its reproduction. Through genetic approaches, we are determining genes and pathways responsible for this response for transfer into potato. B. To develop resistant potatoes through classical breeding methods by breeding with resistant germplasm imported from europe or south america. C. To rear PCN for experimental purposes and to conduct the greenhouse bioassays as part of the steps for DE-regulation of infested fields. 3. Deliverables and expected outcomes: identifying genetic material that confers immunity to PCN that may be transferred into russet-type potatoes. Bioassay samples will be sent to aphis for extraction and aphis will evaluate the data. 4. Intended beneficiary(IES): beneficiaries include owners and operators of PCN infested fields, the potato industry in general and regulatory agencies such as aphis and the idaho state department of agriculture. 5. Subrecipient activities, if known or specified at the time of award: N/a
