Description
- Activities
- Establish real time quaking induced conversion (RT-QUIC) assay for prion detection at scwds for use by cooperative member wildlife agencies for regional research projects and perform inter-laboratory validation of test results. This objective addresses the funding priority to improve the management of CWD-affected wild cervid populations. The expected benefit is to expand CWD diagnostic and research capacity in the southeastern united states. Conduct RT-QUIC to detect CWD prions in tissues collected from 83 deer in a CWD-endemic region of west virginia and compare results to already-completed immunohistochemistry (IHC) testing. This objective addresses the funding priority to improve the management of CWD-affected wild cervid populations. The expected result is to understand differences in prion detection between IHC and RT-QUIC across a variety of tissue types in this particular deer population. Deliverables and expected outcomes: set up a protocol for running RT-QUIC at scwds; run RT-QUIC at scwds on four different tissue types (retropharyngeal lymph nodes, obex, tonsil, and rectoanal mucosa-associated lymphoid tissue) from 84 deer from west virginia. Intended beneficiary: scwds is a state-federal wildlife health research cooperative in the southeastern united states. The resources and expertise available within scwds are shared amongst the partners and are available for wildlife health diagnostic, research, and consultation purposes. RT-QUIC capacity at scwds will directly benefit wildlife professionals in scwds member state wildlife agencies in alabama, arkansas, florida, georgia, kansas, kentucky, louisiana, maryland, mississippi, missouri, nebraska, north carolina, oklahoma, south carolina, tennessee, virginia, and west virginia, as well as the U.S. Fish and wildlife service national wildliferefuge system, and research partners at the U.S. Geological survey and other collaborating academic institutions.
- Subrecipient activities
- DR. Mathison; DR. Mathiason's lab has established and validated RT-quic assays. She will be supporting this project by providing standard, known positive samples for initial assay set-up/validation as well as testing a sub-set of the deer tissues to assess inter-laboratory agreement
