Description
Although other RS strains areendemic and common in the southeastern US, the race 3 biovar 2 (R3BV2)subgroup is not established here. USDA-aphis currently regulates THER3BV2 subgroup as a select agent because its high aggressiveness at cooltemperatures is believed to make IT a unique and serious threat to uspotato production. The primary purpose of this project is to preciselycircumscribe the authentically threatening RS strains and to developdiagnostic markers specific to that population. This will be achievedusing direct phenotypic bioassays of representative RS strains toconfidently determine their level of aggressiveness at cool temperatureson potato. These results will be complemented with in-parallelbioinformatics analyses to evaluate the correlation of variousgenome-wide molecular markers with relevant phenotypic data. A majoranticipated accomplishment of this project will be the generation ofcritical phenotypic data on the aggressiveness of RS strains at wiltingpotato at cool temperatures. Another includes the coupling of thesephenotypic data to genomic markers and the development of PCR assaysthat enable rapid diagnostics, founded in a solid pest-risk analysis.benefits in regard to economic and trade impacts could be significant.improving regulation and diagnostics for select agent RS pathogens canprevent costly, time-consuming problems, and help safeguard the uspotato industry, valued at around $4B per year
