Description
- Purpose
- The purpose of this grant is to develop a web-based tool to enable broader use of AI/ML tools in the design of large protein sequence libraries for measurements of sequence-function relationships.
- Activities
- The researchers will develop a web interface that accepts user inputs for a protein of interest and returns AI/ML model results that use evolutionary information (using sequences of related proteins from nature) to predict the functional impact of sequence changes. The web interface will also output sequences for an oligo pool that can be used to assemble a large set of protein sequences for measurement. The researchers will work with the NIST cellular engineering group to create and measure resulting protein libraries and to characterize the performance of the new library generation web interface and related tools.
- Expected outcomes
- The web interface and related software tools developed by this project will provide an important new capability for use by NIST and other research institutions to generate large protein sequence libraries that have a higher proportion of functional variants which will improve the efficiency with which protein sequence-function measurements can be implemented to learn have changes to sequence result in changes to function.
- Intended beneficiaries
- The web interface developed by this project will benefit NIST programs and will also be accessible to the larger bioscience and biotechnology communities.specifically, this new capability will be applied to the NIST biosecurity/sequences of concern seri project and future related efforts. Also, use of the web interface will not require a high level of AI/ML expertise, so IT will significantly lower the barrier to the generation of high-quality protein sequence-function data both at NIST and at other research institutions.
