Description

In vitro and in vivo experimental evolution of eubacterial rnase p we explore the catalytic versatility and adaptability of the RNA component of a critical ribonucleoprotein, rnase p. Using in vitro evolution, we evolve a descendant of e. Coli rnase p RNA capable of cleaving a DNA substrate. The ability to cleave this substrate involves a tradeoff between catalytic specialization and the ability of rnasep to process multiple RNA substrates, a tradeoff that may be mitigated by the presence of the C5 protein moiety of rnasep. We explore this tradeoff, and its implications for the evolution of novel catalytic activities. We also propose a series of in vivo experiments that result in the DE novo evolution of an rnasep RNA capable of complementing a deletion of the gene (rnpb) encoding the RNA moiety of this ribonucleoprotein in e. Coli. The results will shed light on the topography ofthe adaptive landscape of this ribozyme, and identify the structural elements and sequence motifs critical to catalytic function. Finally, we envision a parallel set of experiments that explore the evolutionary landscape of the protein moiety of rnasep, and that examine the interplay between overall gene expression (the "transcriptome") and the catalytic activity of a single component of metabolism.
