The project will conduct whole-exome and whole-genome sequencing in approximately 450 individuals from a well-characterized tooth agenesis dataset to identify rare variants and test complex inheritance models, then validate findings using whole-genome sequencing data from more than one million individuals across three large biobanks (BioVU, UK Biobank, All of Us) linked to electronic health records. In vitro functional characterization approaches will determine whether identified variants have functional consequences on gene and protein function, and clinical significance of tooth agenesis-associated loci will be explored to improve understanding of genotype-phenotype correlations and support development of diagnostic and regenerative therapies. Work is performed in Pittsburgh, Pennsylvania, with a period of performance extending from September 1, 2026, through May 31, 2031.
