Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
PURCHASE DESCRIPTION
1. Statement of Need and Purpose: The purpose of this acquisition is to provide supplies to the Center for Alzheimer’s Disease and Related Dementias for neurodegenerative disease research for the purpose of long read sequencing on Oxford Nanopore manufactured sequencing systems. These reagents will be used to identify causative genes for neurodegenerative diseases.
2. Background Information and Objective:
Building on National Institute on Aging’s Center for Alzheimer’s Disease and Related Dementias (CARD) existing long-read whole-genome sequencing (WGS) data for The Religious Orders Study and Memory and Aging Project (ROSMAP) brain samples, the lab is extending this work to generate matched long-read RNA sequencing from 250 frontal cortex brain samples to identify transcript-level differences associated with Alzheimer’s disease(AD). The availability of matched long-read genomic, epigenetic, and transcriptomic data from the same individuals provides a uniquely powerful framework to interrogate the genetic, epigenetic, and transcriptional mechanisms underlying AD.
Isoform-resolved transcriptomic profiles will enable detection of disease-associated splicing, expression, and RNA modification changes that are not accessible with short-read approaches. Integrated with the rich pathological and clinical annotations available for ROSMAP, this project will create a high-value, multi-omic long-read resource for the community and establish a scalable model for comprehensive molecular profiling in neurodegenerative disease.
