The research combines the TG344-19 transgenic rat model of Alzheimer's disease with intrastriatal injections of alpha-synuclein preformed fibrils to replicate the full spectrum of dementia with Lewy bodies pathology, including amyloid-beta plaques, Lewy bodies, neurofibrillary tangles, and nigrostriatal degeneration. The R61 phase focuses on developing, optimizing, and internally validating the model to demonstrate cognitive deterioration, Parkinsonian features, rapid eye movement sleep disorder, and visual hallucinations observed in human disease. The resulting model is intended to serve as a platform for preclinical mechanistic and therapeutic studies addressing the limited therapeutic options currently available for dementia with Lewy bodies. Work is performed in East Lansing, Michigan. The period of performance runs through May 31, 2028.
