The National Institute of Neurological Disorders and Stroke awarded the University of Alabama at Birmingham $499,648 on September 1, 2026, under the Extramural Research Programs in the Neurosciences and Neurological Disorders program (CFDA 93.853) to investigate heparan sulfate-mediated mechanisms of alpha-synuclein pathology in dementia with Lewy bodies disease. The project examines how heparan sulfate proteoglycans promote the neuronal internalization of alpha-synuclein fibrils, a pathological hallmark of dementia with Lewy bodies. The research will measure how manipulating heparan sulfate chain length and sulfation patterns impact alpha-synuclein fibril uptake in vitro using cortical neurons from mouse models and in vivo in substantia nigra, hippocampus, and cortex tissue. Pilot data indicates that disrupting electrostatic interactions between heparan sulfate and alpha-synuclein fibrils—potentially through blocking heparan sulfate cell receptors or altering heparan sulfate levels and composition—substantially impairs neuronal internalization of alpha-synuclein fibrils. The work aims to identify therapeutic targets for slowing disease progression and behavioral deficits in synucleinopathy. The award period runs from September 1, 2026, through August 31, 2027. Research is conducted in Birmingham, Alabama. The assistance type is a Project Grant.
