The research team at the University of Virginia will use single-cell and bulk RNA sequencing of mesenchymal stem cells cultured under osteogenic conditions to map eQTLs active during osteogenic differentiation. These context-specific eQTLs will then be integrated with BMD GWAS data to pinpoint candidate genes likely responsible for BMD associations. The researchers will functionally validate these candidate genes using CRISPR-based approaches to assess their role in osteoblast function and bone formation. This work has the potential to uncover novel regulatory mechanisms in bone biology and identify therapeutic targets to treat and prevent osteoporosis.
