This $435,875 five-year Project Grant awarded by the National Institute of General Medical Sciences (NIGMS) Biomedical Research and Research Training Program (CFDA 93.859) supports research to enhance the understanding of epithelial cell polarization and behaviors in tissue morphogenesis. The overarching objectives are to investigate the molecular mechanisms of apical intercalation and apical membrane protein sorting in mammary epithelial cells, using in vitro models and in vivo validation through intraductal injection of genetically manipulated primary cells. The research will employ advanced live cell imaging, super-resolution visualization, soft matter physics, computational modeling, and a new mouse model to analyze the role of actomyosin contractility during the intercalation process and the shaping of mammary ducts. Additionally, the project will investigate the mechanism of apical membrane protein sorting, leveraging synthetic biology approaches, endogenous tagging, and multiplexed super-resolution fluorescence imaging. The findings are expected to provide valuable insights into fundamental processes underlying cell organization and contribute to understanding human diseases.
