The Broad Institute of MIT and Harvard received a $436,250 Project Grant from the National Center for Advancing Translational Sciences (NCATS, CFDA 93.350) awarded May 1, 2026, with completion scheduled for April 30, 2028. Under this award, the Broad Institute is developing and validating the RAPID (Rare Disease Analysis Through Pooled Image-Based Identification of Drugs) platform, a novel massively parallel screening technology designed to accelerate drug repurposing for rare genetic disorders. The platform integrates pooled optical variant identification, high-dimensional image-based profiling, and advanced computational analysis to simultaneously test FDA-approved drugs against hundreds of barcoded disease phenotypes in a single microplate well, dramatically reducing the cost and time required for therapeutic discovery. The deliverables include the development and optimization of the RAPID platform, execution of a pooled repurposing screen testing 220 FDA-approved drugs across 120 diverse rare diseases with previously identified protein mislocalization phenotypes, analysis of 26,400 drug-disease combinations, and confirmation of the most promising therapeutic candidates through dose-response analysis. The Broad Institute will also coordinate with disease-specific experts and disease foundations to advance validated candidates toward clinical development and will make all resulting data and code publicly available through established repositories, supporting the NCATS mission to overcome bottlenecks in the translational research pipeline and accelerate the delivery of therapeutics to patients with rare diseases.Federal Project Grant Award Summary
