This Project Grant award of $222,904.00, provided by the National Institute of Biomedical Imaging and Bioengineering (NIBIB) under the "Discovery and Applied Research for Technological Innovations to Improve Human Health" program (CFDA 93.286), supports the development of a programmable human artificial chromosome (HAC) system for cell-based therapeutics, diagnostics, and screening. The key objectives are to: 1) use the HAC to program inducible expression of multiple therapeutic proteins in response to inflammation, with the aim of treating autoimmune diseases, and 2) build a HAC-based sensor cell line for screening associated with the innate immune response. The award will enable the awardee, President and Fellows of Harvard College (doing business as Harvard Medical School), to rigorously address challenges in robust, stable, and regulated expression of multiple transgenes and genetic circuits within the HAC platform. This research seeks to enhance the ability to explore cells through controlled genetic composition at higher throughput, with applications in disease sensing and therapeutic development.
