This Project Grant award from the National Institute on Aging (CFDA 93.866 - Aging Research) will provide $431,750 to Arizona State University (ASU) to investigate the mechanisms by which a specific genetic variant in the apolipoprotein E (APOE) gene, APOE R145C, increases the risk of Alzheimer's disease (AD) among individuals of African ancestry. The research will utilize ASU's expertise in stem cell bioengineering, neurodegenerative disease modeling, and genome editing to generate human induced pluripotent stem cell lines harboring the APOE R145C variant. Subsequent biochemical, cellular, and genetic analyses of neuronal, astrocytic, and microglial cultures will aim to elucidate how this variant modulates AD-related phenotypes, such as amyloid-beta processing and tau hyperphosphorylation. The findings from this project are expected to inform the development of targeted therapies to address the elevated AD risk in the African American population. The project period will run from September 1, 2024 to June 30, 2026.
