The recipient will conduct basic and translational research examining how iron-dependent cell death mechanisms in gingival keratinocytes contribute to periodontal bone loss. Preliminary data from the recipient demonstrate that keratinocytes in the junctional and sulcular epithelium of patients with periodontitis express ferroptosis features absent in healthy specimens. In a mouse model of ligature-induced periodontitis, increased iron burden accelerated bone loss and upregulated ferroptosis markers in gingival keratinocytes, correlating with increased IL-33 expression and enhanced numbers of ST2+ immunocytes. The work will test whether modulation of glutathione peroxidase 4, an upstream ferroptosis regulator, predisposes gingival keratinocytes to ferroptosis and worsens periodontitis through a mechanism involving release of inflammatory alarmin IL-33. Performance occurs in Philadelphia, Pennsylvania, with a period of performance from July 16, 2026, through April 30, 2031. The assistance type is a Project Grant (R01).
