Award: Mechanisms of Endothelial Cell Differentiation During Vasculogenesis This project grant supports basic research investigating the mechanisms by which endothelial cells (ECs) differentiate and specialize during early vascular development. The research focuses on ETV2, an ETS-family transcription factor that serves as a master regulator of endothelial cell specification. Using human induced pluripotent stem cell (iPSC)-derived mesoderm progenitors and advanced molecular techniques including single-cell RNA sequencing (scRNA-seq), single-cell ATAC sequencing (scATAC-seq), CUT&RUN analysis, and functional CRISPR screening, the project will characterize how ETV2 expression strength and timing influence arteriovenous differentiation and how ETV2 cooperates with the transcriptional repressor REST to suppress alternative mesodermal lineage specification. Deliverables include novel molecular and mechanistic insights into endothelial cell lineage specification and differentiation pathways that advance understanding of vascular development and support future vascular regenerative medicine applications.Federal Project Grant Award Summary
