The recipient will conduct computational discovery of precision therapeutics for hidradenitis suppurativa, a chronic systemic inflammatory skin disorder. Work comprises three aims: identifying immunologic signatures and pathways in HS pathophysiology by analyzing multi-omic datasets of HS and other inflammatory skin conditions; leveraging in silico computational drug repositioning to identify disease-specific and cell-type-specific novel single-agent and combination therapies from existing drugs based on reversal of gene expression signatures; and validating the effects of these agents on skin inflammation using ex vivo skin culture models derived from HS patients, including measuring immune cell activation and proliferation, cytokine production, and characterizing drug-target relationships through structural and computational methods. The work expects to identify novel single and combination treatments that can be repurposed for HS treatment. Performance occurs in San Francisco, California, with a period of performance through August 31, 2031.
