About this file

This statement of work outlines technical requirements for a new hybrid fluorescence microscope for the USDA Agricultural Research Service in Peoria, Illinois. The microscope must have the ability to function as both an upright and inverted system using built-in video monitoring and touchscreen controls. It is required to include a minimum 5MP monochrome camera for fluorescence imaging and 12MP color camera for brightfield imaging, along with fluorescence filter cubes for DAPI and FITC imaging. Up to six objective lenses must be accommodated including 10x, 20x, 40x, and 100x oil objectives. Additional requirements include brightfield, darkfield, phase contrast, and polarized filtering capabilities using a 0.85 NA phase turret condenser. The delivered microscope must not exceed specified dimensions and be delivered by liftgate truck no later than 120 days after contract award. Training is to be provided immediately following installation.

Statement of Work 12505B23Q0220 Hybrid Fluorescence Microscope

General Information

1.0 Scope of Work

Vendor is to deliver and install a new Hybrid (Upright/Inverted) Fluorescence Microscope having the technical requirements as identified in this solicitation.

2.0 Background

Microscopes are typically designed upright systems where the objective lenses are placed above the stage where you put your sample, while inverted microscopes have objective lenses below the sample stage. Requirements for each design are dependent on the sample type that is being viewed. As such, laboratories will often have both types of microscopes available. The USDA Agricultural Research Service (ARS) in Peoria, Illinois has a need to utilize a single microscope with built-in video imaging/capturing that can perform as either an inverted or upright system to minimize required laboratory space. In addition, this microscope must have brightfield, darkfield, fluorescence, phase-contrast, and polarized filtering imaging capabilities.

Contractor Requirements

3.0 Technical Requirements / Tasks

The new hybrid fluorescence microscope must have the following technical requirements:

1. Ability to be used in upright format (i.e., objective lenses below sample stage) for samples in slides/coverslips.

2. Ability to be used in inverted format (i.e., objective lenses above sample stage) for samples in petri dishes, microplates, and flasks.

3. Built-in video monitoring with touchscreen functions to control cameras and illumination.

4. Ability to capture imaging with a minimum 5 MP CMOS monochrome camera for fluorescence imagine and 12 MP color camera for brightfield imaging.

5. Ability to perform fluorescence imaging with up to four cubes, with the following included in the system:

a. DAPI - EX:380/30 EM:450/50 DM:425

b. FITC - EX:470/40 EM:525/50 DM:495

6. Ability to accommodate up to six objective lenses, with the following objectives or better included in the system:

a. 10x Plan Fluorite Phase, NA 0.30 WD 10mm

b. 20x Plan Fluorite Phase, NA 0.50 WD 2.1mm

c. 40x Plan Fluorite Phase, NA 0.75 WD 0.51mm

d. 100x Plan Fluorite Oil Phase, WD 0.2mm NA 1.30

7. Ability to perform brightfield, darkfield, phase contrast, and polarized filtering with a Phase Turret Condenser having a numerical aperture of 0.85 (WD 7mm) or better.

8. Ability to hold glass slides on sample stage.

9. Ability to perform optical Z-stack function for automated acquisition of multiple focal planes.

10. Ability to create panoramic image stitching.

11. Ability to save images and associated data to USB drive.

12. Size must not exceed 24” depth, 15” width, and 20” height.

4.0 Deliverables / Schedule

This instrument will need to delivered to 1815 N University St. Peoria, Illinois 61604. Delivery will require the use of a liftgate.
