1.0 PRICE SCHEDULE
The USDA, Animal and Plant Health Inspection Service (APHIS), Plant Protection and Quarantine (PPQ), Plum Pox Virus (PPV) Eradication Program, has a requirement for diagnostic laboratory services to process government-supplied leaf samples potentially infected with the Plum Pox Virus. Samples will be collected in New York State during the months of May through October. The actual days of collection depends on the season and condition of the leaves. Leaf samples will consist of leaves taken from commercial orchards, nurseries, residential properties and wild areas in the northwestern region of New York State.
The diagnostic laboratory contractor shall provide a price per sample for the range of samples listed below. Quantities vary each year. The price per sample shall include all costs incurred by the contractor laboratory providing the services requested. The contractor receiving the award must be able to handle the volume of a few hundred to several thousand samples arriving per day, depending on the conditions of season.
The number of samples processed in the previous contract were approximately 125,000 in 2013, 97,000 in 2014, 70,000 in 2015, and 45,000 in 2016—the bulk of which came in June (9,900), July (19,350), and August (15,950). Samples to be processed in 2017 are estimated to be 50,000. The number of samples to be collected in 2018 and beyond will be dependent on identification of positive samples which would determine the area of survey and number of samples to collect for the succeeding year. If positive samples are identified 2017, the number of samples to be collected in 2018 may remain at a similar sample size as 2017. If no positive samples are identified in 2017, the number of samples to be collected in 2018 will in all likelihood decrease. The number and location of positive PPV samples will determine the final sample number for all succeeding years.
As a result of this solicitation, the government intends to award a four-year, commercial, indefinite-delivery indefinite-quantity (IDIQ) contract. As such, the guaranteed minimum order for the resultant contract is 1,000 samples. The contract maximum is 250,000 samples. There is no guarantee, however, that orders above the guaranteed minimum will be ordered throughout the remaining life of the contract.

2.0 STATEMENT OF WORK
A. Purpose:
The purpose of diagnostic laboratory services is to receive and process New York State Prunus leaf samples to accurately detect the presence of the Plum Pox Virus (PPV).
B. Processing Samples:
1. See Attachment A for the protocol used for USDA regulatory testing of PPV.
2. The testing laboratory shall use the most current version of the CPHST-specified ELISA Work Instruction B-T-4-3. Any changes or updates to the current protocol will be provided:
https://www.aphis.usda.gov/aphis/ourfocus/planthealth/plant-pest-and-disease-programs/pests-and-diseases/plum-pox/ct_plum_pox
3. A lab shall a have a minimum bio-security level (BSL) 1 clearance. A BSL 1 lab is defined as follows, per USDA DM 9610-001: Practices, safety equipment, and facility design and construction are appropriate for undergraduate and secondary educational training and teaching laboratories, and for other laboratories in which work is done with defined and characterized strains of viable microorganisms not known to consistently cause disease in healthy adult humans.
4. Leaf Samples https://www.aphis.usda.gov/aphis/ourfocus/planthealth/plant-pest-and-disease-programs/pests-and-diseases/plum-pox/ct_plum_pox https://www.aphis.usda.gov/aphis/ourfocus/planthealth/plant-pest-and-disease-programs/pests-and-diseases/plum-pox/ct_plum_pox
a. Field samples must be stored at 4°C and shall be processed as soon as possible.
b. Samples that are partially decayed (have a brown appearance, are water soaked, or moldy) shall not be tested due to potentially decreased absorbance values. These materials must be recollected.
c. In cases when re-testing is required processing up to two weeks after collection can still take place as long as leaves are stored properly (4oC) and remain in good condition.
Longer storage will decrease the amount of virus detected due to degradation of the virus in the aging plant tissue that can result in false negative results.
d. Samples for PPV shall be kept cold (4°C) for virus integrity; leaf samples shall not be stored in a freezer.
e. If plant samples arrive warm to the touch, they shall be placed in a 4°C refrigerator for several hours prior to handling. Experience has shown that the titer will be diminished if samples are not first chilled to stop degradation.
f. Gloves shall be worn when handling samples and should be changed between samples.
5. Sample Testing
a. Any survey sample that tests positive by ELISA shall be delivered to the Federal Confirmatory laboratory without additional testing by the Contractor laboratory.
b. A PPQ-391 form must be sent to the Federal Confirmatory Lab along with every PPV suspect sample(s) submitted for testing. The Federal Confirmatory Lab will not begin testing unless a 391 form is on file.
c. The Contractor will be notified by the APHIS-PPQ-PPV Eradication Program (not the Federal confirmatory laboratory) if any of the samples submitted test inconclusive by the federal confirmatory lab. If so, arrangements shall be made, with the APHIS-PPQ- PPV Eradication Program, for sample recollection by authorized personnel as soon as possible. The newly collected sample(s) shall be forwarded for federal confirmation
d. All negative samples processed by the Contractor laboratory are considered negative for USDA-PPQ regulatory purposes.
6. Leaf processing time
a. Samples will be received four days per week, Tuesday – Friday.
b. The number of samples received shall equal the number of samples processed by the laboratory.
c. In order to prevent the accumulation of unprocessed samples at the lab, the laboratory shall work closely with the USDA to prevent unprocessed sample accumulation. If more than three days of unprocessed samples accumulate in the lab, USDA shall be notified.
C. Suspect sample instructions:
1. Samples testing positive for PPV using the PPQ, CPHST-authorized ELISA screening test shall be uniquely identified and secured for shipment to the USDA Confirmatory Laboratory. The following USDA personnel shall be notified by email that a PPV ELISA positive sample has been identified and that shipment to the USDA Confirmatory Laboratory is expected:
a. APHIS-PPQCPHSTBeltsvilleSampleDiagnostics@aphis.usda.gov and PPQ.Domestic.Diagnostic.Coordinator@aphis.usda.gov
b. The email should also be copied to the following PPV Program and State personnel:
mailto:APHIS-PPQCPHSTBeltsvilleSampleDiagnostics@aphis.usda.gov mailto:PPQ.Domestic.Diagnostic.Coordinator@aphis.usda.gov
i. Current SPHD
ii. Current SPRO

3. The CPHST Beltsville laboratory shall email the Contractor laboratory immediately upon receiving the samples only if there is a problem with the samples. This email should also be copied to the current PPV Program Director, the current CPHST Beltsville Laboratory Director and the current CPHST Beltsville Laboratory Manager.
4. The Contractor laboratory shall review feedback and arrange for re-sampling of the rejected samples as soon as possible. Re-samples should be sent directly to CPHST Beltsville laboratory for testing using above mentioned protocols (Step 1 – 3)
5. The CPHST Beltsville laboratory shall report test results of PPV suspects to the NIS Domestic
Diagnostic Coordinator who will disseminate the information to the PPV Program Director, New York State Plant Health Director (SPHD), New York State Plant Regulatory Official (SPRO) and the CPHST National Science Director (NSD).
6. The SPHD and PPV Program Director shall inform the Contractor laboratory supervisor(s) of test results and request resample if required.
D. Proficiency Testing:
The Contractor laboratory shall successfully complete a USDA-PPQ PPV Proficiency Test administered by the National Plant Pathogen Laboratory Accreditation Program (NPPLAP) each season prior to ELISA testing of any PPV survey samples. This program is described on the PPQ NPPLAP website. The Contractor shall contact the current NPPLAP Coordinator to discuss the NPPLAP process for proficiency tests. The Contracting laboratory personnel directly performing the PPV ELISA testing, or directly responsible for ELISA testing for PPV (as determined by the Contractor and USDA-PPQ), shall complete the PPV-specific proficiency test distributed by NPPLAP before beginning sample testing.
