GENERAL INFORMATION

Title: Labconco® Acid Resistant CentriVap Pro Configuration (Centrivap, cold-trap, and vacuum pump)

1. Background The Laboratory of Genome Integrity and Structural Biology at the National Institute of Environmental Health Sciences (NIEHS) conducts research to understand the molecular mechanisms that maintain genome integrity and how environmental exposures contribute to DNA damage, genome instability, and cellular stress responses.

Current research activities include metabolomics analyses and purification and characterization of DNA adducts for downstream analytical measurements. These research workflows require the preparation, concentration, and drying of samples containing aqueous, organic, acidic, and other chemically challenging solvents. Reliable concentration and drying of these samples is necessary to preserve sample integrity and prepare materials for subsequent analyses, including liquid chromatography-mass spectrometry (LC-MS).

2. Objective The objective of this acquisition is to obtain a Labconco® Acid-Resistant CentriVap Pro configuration, consisting of the CentriVap concentrator, cold trap, and compatible vacuum pump, for use in metabolomics sample preparation and DNA adduct purification workflows.

The system must provide the chemical resistance, controlled evaporation, cold-trap protection, vacuum performance, and sample capacity necessary to safely and reproducibly process research samples containing acidic, aqueous, organic, volatile, and mixed solvents.

3. Scope of Work The contractor shall provide a complete, operational Labconco Acid-Resistant CentriVap Pro configuration, including all components and accessories necessary for normal operation.

The required configuration shall include, at a minimum:

· Labconco Acid-Resistant CentriVap Pro vacuum concentrator.

· PTFE-coated chamber suitable for samples prepared under acidic or chemically harsh conditions.

· DNA PTFE rotor, Labconco catalog number 7555701.

· Labconco −105°C CentriVap cold trap, including stainless-steel lid and 115V North American electrical configuration, catalog number 8621100.

· Compatible Labconco VACUUBRAND diaphragm vacuum pump, configured for 115V/60Hz operation, with an ultimate vacuum below 2 mbar and displacement capacity of approximately 63 L/min.

· All required tubing, connections, electrical components, and accessories necessary to integrate the CentriVap, cold trap, and vacuum pump into a functional system.

4. Essential Technical Requirements

4.1 Acid and Chemical Resistance

The concentrator must be suitable for samples containing chemically harsh or corrosive solvents and reagents encountered in DNA adduct purification and metabolomics workflows.

The system shall incorporate:

· A PTFE-coated aluminum chamber to provide chemical resistance.

· A chemical-resistant vapor path suitable for materials including, but not limited to, trifluoroacetic acid (TFA), hydrochloric acid, acetic acid, ammonium hydroxide, and sodium hydroxide.

· A glass lid and phenol-free lid gasket to minimize chemical interaction and potential sample contamination.

4.2 Controlled Evaporation

The system shall provide controlled evaporation conditions appropriate for sensitive biological and chemical samples.

The concentrator shall provide:

· Controlled heating from ambient temperature to approximately 100°C.

· Independent programming of heating and total run time.

· The ability to terminate heating before completion of the vacuum run to minimize unnecessary thermal exposure of labile metabolites and DNA adducts.

4.3 Cold-Trap Protection

The system shall include a −105°C cold trap capable of capturing moisture, solvent vapors, and corrosive fumes generated during sample concentration.

The cold trap is required to:

· Protect the vacuum pump from moisture and chemical vapors.

· Provide appropriate vapor capture for volatile and low-freezing-point solvents.

· Support LC-MS sample preparation workflows involving volatile organic solvents.

· Reduce the potential for corrosive or solvent vapors to enter the vacuum pump.

4.4 Rotor and Sample Capacity

The system shall include a DNA PTFE rotor suitable for small-volume DNA adduct and metabolomics sample processing.

The configuration shall:

· Support the required DNA PTFE rotor format.

· Accommodate sample volumes up to approximately 50 mL, as appropriate for the specified configuration.

· Support concentration of small-volume samples prior to downstream analytical procedures, including LC-MS.

4.5 Vacuum Pump and Integration

The vacuum pump shall be compatible with the CentriVap system and suitable for aqueous, volatile, low-boiling-point, and mixed-solvent applications.

The required pump configuration shall provide:

· 115V/60Hz electrical operation.

· Ultimate vacuum below approximately 2 mbar.

· Displacement capacity of approximately 63 L/min.

· Chemical-resistant wetted components suitable for the intended solvent applications.

· Compatibility with the CentriVap's integrated controls.

The CentriVap configuration shall provide an electrical receptacle or equivalent integrated control capability allowing the vacuum pump to operate through the concentrator controls, including use of the system's built-in vacuum delay functionality.

5. Research Applications The system will support NIEHS research activities requiring concentration and drying of samples for metabolomics and DNA adduct analyses. These workflows may involve chemically challenging extraction and purification conditions and require careful control of temperature and solvent evaporation.

The requested configuration will provide the capability to:

· Concentrate metabolomics extracts prior to downstream analysis.

· Prepare and dry DNA adduct samples for subsequent measurements.

· Process samples containing aqueous, organic, acidic, and mixed solvents.

· Minimize sample loss and degradation during concentration.

· Protect vacuum equipment from corrosive and volatile vapors.

· Provide reproducible sample preparation conditions across experiments.

6. Government Quality Assurance Government acceptance and quality assurance will be performed following delivery and installation of the equipment. The Government will verify that the delivered system conforms to the required configuration and includes the specified major components and accessories.

Acceptance testing will include, as applicable:

· Verification of the Labconco Acid-Resistant CentriVap Pro configuration.

· Verification of the DNA PTFE rotor, catalog number 7555701.

· Verification of the −105°C CentriVap cold trap, catalog number 8621100.

· Verification of the compatible 115V diaphragm vacuum pump.

· Verification of proper system integration and electrical operation.

· Verification of appropriate vacuum generation and operation.

· Verification of cold-trap operation and temperature performance.

· Verification of rotor installation and compatibility.

· Demonstration of controlled heating and concentration functions using standard laboratory procedures.

The delivered system shall be operational and capable of performing the intended sample concentration and drying functions prior to Government acceptance.

7. Justification of Configuration The requested Labconco Acid-Resistant CentriVap Pro configuration is specifically suited to the Laboratory of Genome Integrity and Structural Biology's metabolomics and DNA adduct purification workflows because it combines acid-resistant construction, controlled evaporation, low-temperature cold trapping, appropriate rotor capacity, and chemical-resistant vacuum pumping in an integrated system.

These capabilities are necessary to accommodate the range of solvents and sample preparation conditions used in the laboratory while protecting valuable research samples and associated vacuum equipment. The combination of the specified components provides a complete system designed for the laboratory's intended applications rather than requiring separate equipment or modifications to accommodate individual sample preparation requirements.

8. Deliverable The contractor shall deliver a complete, new, operational Labconco Acid-Resistant CentriVap Pro configuration, including the specified CentriVap, DNA PTFE rotor, −105°C cold trap, compatible diaphragm vacuum pump, and all required accessories and components necessary for integrated operation.

The equipment shall be delivered in accordance with the manufacturer's specifications and shall be ready for use in NIEHS research applications following installation and verification.

Period of Performance: This is a one-time supply acquisition with the duration of the maintenance agreement as specified in the resulting contract or purchase order.

Delivery Location:
