Aim 2 engineers osteoclast-targeting nanoparticles functionalized to deliver RANK siRNA, suppress osteoclastogenesis, and prevent alveolar bone resorption. Aim 3 establishes proof-of-concept for nanoparticle-enabled CAR-macrophages that recognize and eliminate osteoclasts, reprogram the local microenvironment, and provide sustained immunomodulation. Upon completion, the work will establish a scalable nanoparticle platform for precision delivery of therapeutics to inflammatory and bone-resorptive pathways in periodontal disease.
