DECIPHER-OCD: Comprehensive Characterization of Brain Structure in Obsessive-Compulsive Disorder Yale University has been awarded $460,625 under the National Institute of Mental Health (NIMH) Mental Health Research Grants program (CFDA 93.242) for a two-year project period from August 1, 2026 through July 31, 2028. The project delivers a comprehensive neuroimaging analysis pipeline designed to systematically characterize brain structural abnormalities in obsessive-compulsive disorder (OCD) using advanced morphometric measures. The research team will apply this pipeline to structural magnetic resonance imaging data from 2,274 OCD cases and 2,280 control subjects collected across 26 international sites as part of the ENIGMA-OCD Consortium, assessing multiple structural metrics including cortical thickness, surface area, regional volume, five curvature indices, and novel brain network-derived measures (MIND and CSMIND) not previously evaluated in OCD populations. The project addresses significant knowledge gaps by moving beyond conventional structural measures to explore curvature metrics and network-derived indices that pilot analyses suggest may provide enhanced insight into OCD-associated circuit abnormalities. Using canonical correlation analysis and other novel statistical approaches, the research will examine biological correlates of spatial variation patterns associated with OCD diagnosis, generating findings intended to advance understanding of OCD neurobiology and inform development of new treatment strategies for this prevalent, frequently chronic disorder linked to elevated mortality and substantial functional impairment.
