Project Grant R21CA318125
- The National Cancer Institute awarded Case Western Reserve University $100,228 on December 1, 2026, under the Cancer Research Manpower program (CFDA 93.398) to develop real-time monitoring of nanobubble-labeled chimeric antigen receptor T cell trafficking via contrast-enhanced ultrasound imaging. The award funds research to refine nanobubble labeling protocols for prostate-specific membrane antigen (PSMA)-targeting CAR-T cells, enhancing imaging duration and stability to enable extended...
- The National Cancer Institute awarded $413,339 to Cleveland Clinic Lerner College of Medicine of Case Western Reserve University on August 1, 2026, under the Cancer Treatment Research program (CFDA 93.395) to conduct research on chimeric antigen receptor (CAR) T-cell therapy for lymphoma treatment. The research aligns T-cell biology with CAR architecture to enhance lymphoma therapy, addressing the limitation that durable responses are achieved in only 35–50% of lymphoma patients currently...
- The National Cancer Institute, part of the Department of Health and Human Services, awarded Case Western Reserve University $630,108 on March 13, 2026, under the Cancer Detection and Diagnosis Research program (CFDA 93.394). The recipient will develop and evaluate theranostic approaches to breast cancer using a combined fluorescence image-guided surgery and photothermal therapy platform centered on AKRO-6QC, a molecule designed for specific binding and activation by cysteine proteases...
- The National Cancer Institute awarded Case Western Reserve University $164,190 under the Cancer Research Manpower program (CFDA 93.398) on January 16, 2026, to investigate the role of B-cell activating factor (BAFF) in adverse events associated with chimeric antigen receptor (CAR)-T cell therapy. The research examines BAFF's contribution to cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS), the two most common and potentially life-threatening...
- This Project Grant award from the National Cancer Institute (CFDA 93.395 - Cancer Treatment Research) is providing $827,942.00 to the Cleveland Clinic Lerner College of Medicine of Case Western Reserve University to investigate a novel inhibitory receptor that regulates the persistence and function of chimeric antigen receptor (CAR)-engineered T cell therapy for lymphoma. The key objectives are to: 1) examine the causal role of this inhibitory receptor axis in impairing the therapeutic...
- The National Cancer Institute awarded Case Western Reserve University $137,890 under the Cancer Research Manpower program (CFDA 93.398) on December 1, 2025. The award funds development of a deep learning computational imaging model to identify rectal cancer patients who achieve complete response to total neoadjuvant therapy and are candidates for watch-and-wait treatment approaches. The model will use specialized image filters to quantitatively characterize subtle desmoplastic stromal...
- The Department of Defense's Military Medical Research and Development program (CFDA 12.420) awarded Case Western Reserve University's Office of Research Administration Division, doing business as University Hospitals of Cleveland, a $1,467,210 Project Grant on September 30, 2024. The grant supports research to develop a novel dendritic cell therapy approach for mucosal melanoma, focusing on targeting a differentiation epitope in CD11b of tumor-associated inflammatory monocytes. The research aims...
- The National Cancer Institute awarded a Cooperative Agreement worth $667,575 to Case Western Reserve University on July 8, 2026, under the Cancer Biology Research program (CFDA 93.396) to investigate mechanisms by which cancer extracellular vesicles drive breast cancer metastasis and to evaluate therapeutic interventions targeting this pathway. The research will examine the role of kindlin-2 (K2), an oncogenic protein enriched in small extracellular vesicles (SEVs) secreted by breast cancer...
- The National Cancer Institute awarded Cleveland Clinic Lerner College of Medicine of Case Western Reserve University $640,775 on June 22, 2026, under the Cancer Treatment Research program (CFDA 93.395) to conduct a co-clinical trial investigating mechanisms by which intratumoral bacteria drive immunosuppression and immune checkpoint inhibitor resistance in head and neck squamous cell carcinoma. The recipient will define how intratumoral bacterial load modulates the immunosuppressive tumor immune...
- The National Institutes of Health National Cancer Institute awarded The Washington University $399,829 under the Cancer Treatment Research program (CFDA 93.395) on April 1, 2026, to conduct rational design of next-generation engineered T cells for enhanced cancer immunotherapy. The research addresses limited persistence of engineered T cells in cancer treatment by harnessing 4-1BB (CD137/TNFRSF9), a co-stimulatory molecule, through structure-informed protein design. Current 4-1BB chimeric...
The National Cancer Institute, part of the Department of Health and Human Services, awarded Case Western Reserve University $393,812 on August 1, 2026, under the Cancer Treatment Research program (CFDA 93.395) to develop and optimize photobiomodulation (PBM)–enhanced CAR-T cell therapy. The project investigates whether visible to near-infrared light can enhance the metabolic fitness and durability of chimeric antigen receptor T cells without genetic modification. Aim 1 systematically optimizes PBM parameters—wavelength, fluence, and treatment schedule—to maximize CAR-T cell proliferation, cytotoxicity, and mitochondrial fitness in vitro, using metabolic assays including Seahorse analysis, ATP and reactive oxygen species quantification, and expression markers for mitochondrial biogenesis. Aim 2 evaluates whether PBM-treated CAR-T cells improve therapeutic efficacy in vivo using a mammalian rodent model of B-cell lymphoma, measuring tumor burden by bioluminescence imaging, survival, and CAR-T persistence and exhaustion markers. Case Western Reserve University's Office of Research Administration, operating in affiliation with University Hospitals of Cleveland, serves as the recipient. Performance occurs in Cleveland, Ohio through July 31, 2028. This is a Project Grant assistance type.
