The National Cancer Institute awarded a $410,933 Project Grant (Cancer Treatment Research, CFDA 93.395) to the University of California, San Diego, effective March 18, 2026, through February 29, 2028. This exploratory research initiative (R21 mechanism) aims to develop next-generation small interfering RNA (siRNA) therapeutics to treat metastatic castration-resistant prostate cancer (mCRPC). The project focuses on enhancing siRNA delivery into mCRPC tumor cells by approximately tenfold through innovative technology development, enabling these therapeutics to effectively target all genetic forms of the androgen receptor (AR)—the primary oncogenic driver in mCRPC—while minimizing harm to surrounding normal cells. The research deliverables encompass the design and validation of improved siRNA triggers with enhanced cellular uptake capability, leveraging RNA interference (RNAi) mechanisms to selectively degrade oncogenic AR gene products. This work addresses a critical clinical gap, as current anti-androgen drugs become ineffective once the AR gene mutates or becomes amplified in mCRPC patients. By developing siRNA therapeutics capable of targeting all AR oncogenic variants driving tumor growth and therapeutic resistance, the project seeks to provide viable treatment options for an otherwise untreatable lethal disease with significant bone metastasis complications. Testing will be conducted at UCSD's La Jolla, California facility.Federal Grant Award Summary
