This Project Grant award from the National Institutes of Health (NIH) under the Trans-NIH Research Support program (CFDA 93.310) provides $157,500.00 to the University of California, Los Angeles (UCLA) to develop and test new radiation mitigator compounds based on the estrogen receptor beta (ER-beta) agonist DPN. The goal is to create orally bioavailable DPN analogues that can more effectively treat bone marrow, gastrointestinal, respiratory, and cardiovascular injuries caused by total-body irradiation, which are major determinants of mortality after radiation exposure. The research plan involves synthesizing DPN analogues, evaluating their radiation mitigating properties in mouse models, and assessing their mechanisms of action through cytokine and histopathological analyses. This work aims to advance a new class of oral radiation mitigators that could be used in the event of nuclear accidents or attacks, as well as to manage side effects of clinical radiotherapy. The award period runs from August 1, 2025 to July 31, 2027.
