This Project Grant award, totaling $213,000.00, was provided by the National Institute on Alcohol Abuse and Alcoholism (NIAAA) under the Alcohol Research Programs (CFDA 93.273) federal grant program. The goal of this 24-month project, with a period of performance from August 22, 2024 to July 31, 2026, is to assess the role of nitric oxide synthase 2 (NOS2) and the interleukin-22 (IL-22)-STAT3 signaling pathway in alcohol-mediated exacerbation of ulcerative colitis (UC) pathology. The research will be conducted by Loyola University of Chicago's Health Sciences Campus in Maywood, Illinois. The project aims to determine whether alcohol-induced increases in NOS2 expression directly impact UC pathology or do so through bacterial dysbiosis, as well as investigate whether IL-22 utilizes STAT3 to mitigate the upregulation of the NOS2 pathway and prevent alcohol-induced exacerbation of UC in a mouse model. The findings are expected to provide valuable insights into the mechanisms by which alcohol exacerbates UC flare periods, which could inform the development of more effective treatments for these patients.
