This Project Grant award of $157,437 from the National Institute of Environmental Health Sciences (NIEHS) under the Environmental Health (CFDA 93.113) program aims to define the urinary, biliary, and fecal excretion of per- and polyfluoroalkyl substances (PFAS) and the impact of dietary fiber. The key products or services to be delivered under this award include: Analyzing samples collected from humanized PPARA mice exposed to seven common PFAS in drinking water and fed different diets, including those supplemented with dietary fibers and cholestyramine, to establish proof of principle that enterohepatic recirculation is critical to maintaining high PFAS body burdens. Defining the effect of PFAS on transporter expression that could modify PFAS toxicokinetics. The coordinated analyses under this R03 grant and a related TERP grant are expected to generate the toxicokinetic, physiological, and molecular evidence needed to clarify the role of enterohepatic recirculation in PFAS toxicity. The award period runs from May 10, 2025 to May 9, 2027.
