The National Institute of Diabetes and Digestive and Kidney Diseases awarded The Children's Hospital of Philadelphia $699,999 on August 15, 2026, for basic research on chromatin remodelers in erythropoiesis and hemoglobin regulation under the Diabetes, Digestive, and Kidney Diseases Extramural Research program (CFDA 93.847). The funded work dissects molecular mechanisms by which BAF (Brahma-Associated Factor) chromatin remodeling complex subtypes and subunits achieve gene-specific regulation during erythroid development, with direct application to hemoglobin disorders including sickle cell disease and beta-thalassemia. Preliminary data indicate that BAF complexes display selective functions at the beta-hemoglobin gene locus; loss of non-canonical BAF components preferentially reduces expression of fetal and minor adult globin genes while sparing the major adult beta-globin gene. The three research aims include dissecting the scope and mechanisms of non-canonical BAF function in erythroid cells using acute loss-of-function approaches, extending studies in vivo through knockout of the BAF subunit BRD9 in the mouse erythroid lineage, and conducting follow-up experimental studies to achieve gene specificity within the globin locus. Performance is located in Philadelphia, Pennsylvania 19104. The award period runs through April 30, 2031. The Children's Hospital of Philadelphia Research Institute, operating as the Children's Hospital of Philadelphia Foundation, is the recipient.
