The National Institute of Diabetes and Digestive and Kidney Diseases awarded The Research Foundation For The State University Of New York (operating as Stony Brook University) $785,689 on August 1, 2026, under the Diabetes, Digestive, and Kidney Diseases Extramural Research program (CFDA 93.847) to identify the therapeutic potential and molecular mechanism of the long noncoding RNA HOXB3OS in autosomal dominant polycystic kidney disease. The research addresses the incompletely understood pathophysiology of ADPKD, a disease affecting over 600,000 people in the United States, where approximately half of those diagnosed progress to end-stage renal failure requiring dialysis or kidney transplantation. Current FDA-approved treatment is limited to tolvaptan. The recipient will conduct fundamental studies to identify therapeutically targetable long noncoding RNAs that prevent or mitigate cyst formation and progression in ADPKD. Prior work identified HOXB3OS as the first long noncoding RNA directly involved in mouse polycystic kidney disease; the human ortholog HOXB-AS1 is similarly decreased in human ADPKD kidneys and cystic kidney organoids. The research will investigate how HOXB3OS RNA associates with multiple proteins central to ADPKD metabolism, including PKM2 and ATP synthase, with the goal of elucidating new drug targets. Work is performed in Stony Brook, New York. The award period runs from August 1, 2026, through May 31, 2030. This is a Project Grant, the standard assistance type for NIH research.
